TL;DR: It is found that under conditions of stress, such as malignant transformation fuelled by MYC, the chaperome becomes biochemically ‘rewired’ to form a network of stable, survival-facilitating, high-molecular-weight complexes.
Journal ArticleDOI
Adapting to stress - chaperome networks in cancer.
TL;DR: It is argued that in cancer cells, a state of chaperomes hyperconnectivity is obtained by increasing the interaction strength among chaperome machinery members, and this concept will change how drugs targeting the chaperite to treat cancer are developed and implemented.
Journal ArticleDOI
Gold/alpha-lactalbumin nanoprobes for the imaging and treatment of breast cancer.
Jiang Yang,Jiang Yang,Tai Wang,Lina Zhao,Vinagolu K. Rajasekhar,Suhasini Joshi,Chrysafis Andreou,Suchetan Pal,Hsiao-Ting Hsu,Hanwen Zhang,Ivan J. Cohen,Ruimin Huang,Ronald C. Hendrickson,Matthew M. Miele,Wenbo Pei,Matthew Brendel,John H. Healey,Gabriela Chiosis,Gabriela Chiosis,Moritz F. Kircher +19 more
TL;DR: Renally clearable gold quantum clusters that are stabilized by the milk metalloprotein alpha-lactalbumin and display multicolour fluorescence aid the detection, resection and treatment of breast cancer in mice.
Journal ArticleDOI
The epichaperome is a mediator of toxic hippocampal stress and leads to protein connectivity-based dysfunction.

TL;DR: This work uses cellular and animal models as well as human biospecimens to show that AD-related stressors mediate global disturbances in dynamic intra- and inter-neuronal networks through pathologic rewiring of the chaperome system into epichaperomes.
Journal ArticleDOI
Chaperome heterogeneity and its implications for cancer study and treatment
Tai Wang,Anna Rodina,Mark Dunphy,Adriana D. Corben,Shanu Modi,Monica L. Guzman,Daniel T. Gewirth,Gabriela Chiosis +7 more
TL;DR: How ligand–protein binding kinetics are more appropriate than equilibrium binding parameters to characterize and unravel chaperome targeting in cancer and to gauge the selectivity of ligands for specific tumor-associated chaperomes pools is discussed.
