Somatic mutations in ATP1A1 and CACNA1D underlie a common subtype of adrenal hypertension.
Elena A.B. Azizan,Hanne Poulsen,Petronel Tuluc,Junhua Zhou,Michael V. Clausen,Andreas Lieb,Carmela Maniero,Sumedha Garg,Elena G. Bochukova,Wanfeng Zhao,Lalarukh Haris Shaikh,Cheryl A. Brighton,Ada E. Brown +32 more
TL;DR: Exome sequencing of zona glomerulosa–like APAs identifies nine with somatic mutations in either ATP1A1, encoding the Na+/K+ ATPase α1 subunit, or CACNA1D, encoding Cav1.3, which caused inward leak currents under physiological conditions and induced a shift of voltage-dependent gating to more negative voltages, suppressed inactivation or increased currents.
Journal ArticleDOI
Study Heterogeneity and Estimation of Prevalence of Primary Aldosteronism: A Systematic Review and Meta-Regression Analysis
Sabine C Käyser,Tanja Dekkers,H. Groenewoud,Gert Jan van der Wilt,J. Carel Bakx,Mark van der Wel,Ad R. M. M. Hermus,Jacques W.M. Lenders,Jaap Deinum +8 more
TL;DR: It is demonstrated that it is pointless to claim low or high prevalence of primary aldosteronism based on published reports, and the findings urge for a prevalence study whose design takes into account the factors identified in the meta-regression analysis.
Journal ArticleDOI
Adrenal vein sampling versus CT scan to determine treatment in primary aldosteronism: an outcome-based randomised diagnostic trial
Tanja Dekkers,Aleksander Prejbisz,Leo J. Schultze Kool,Hans Groenewoud,Marieke Velema,Wilko Spiering,Sylwia Kołodziejczyk-Kruk,Mark J. Arntz,Jacek Kądziela,Johannes F Langenhuijsen,Michiel N. Lenders,Jacques W.M. Lenders,Jaap Deinum +21 more
TL;DR: A non-significant mean difference of 0·05 (95% CI -0·04 to 0·13) in quality-adjusted life-years (QALYs) was found to the advantage of the AVS group, associated with a significant increase in mean health-care costs of €2285 per patient.
Journal ArticleDOI
Steroid metabolome analysis reveals prevalent glucocorticoid excess in primary aldosteronism
Wiebke Arlt,Katharina Lang,Alice J Sitch,Anna Dietz,Yara Rhayem,Irina Bancos,Annette Feuchtinger,Vasileios Chortis,Lorna C Gilligan,Philippe Ludwig,Anna Riester,Evelyn Asbach,Beverly A. Hughes,Donna M. O'Neil,Martin Bidlingmaier,Jeremy W. Tomlinson,Zaki Hassan-Smith,D. Aled Rees,Christian Adolf,Stefanie Hahner,Marcus Quinkler,Tanja Dekkers,Jaap Deinum,Michael Biehl,Brian G. Keevil,Cedric H. L. Shackleton,Jonathan J Deeks,Axel Walch,Felix Beuschlein,Martin Reincke +29 more
TL;DR: It is indicated that glucocorticoid cosecretion is frequently found in primary aldosteronism and contributes to associated metabolic risk, and mineralocortioid receptor antagonist therapy alone may not be sufficient to counteract adverse metabolic risk in medically treated patients with primary aldononism.
Journal ArticleDOI
An LC-MS/MS method for steroid profiling during adrenal venous sampling for investigation of primary aldosteronism.
Mirko Peitzsch,Tanja Dekkers,Matthias Haase,Fred C.G.J. Sweep,Ivo Quack,Gerald Antoch,Gabriele Siegert,Jacques W.M. Lenders,Jaap Deinum,Holger S. Willenberg,Graeme Eisenhofer +10 more
TL;DR: The LC-MS/MS assay offers profiling of 13 other adrenal steroids, providing a potentially useful method for the clinical work-up of patients with primary aldosteronism, and larger AV/pV ratios of several steroids compared to cortisol suggest more sensitive alternatives to the latter for assessing positioning of AV sampling catheters.
