Journal ArticleDOI
The 5′ and 3′ Untranslated Regions of the Flaviviral Genome
TL;DR: The highly structured 3′UTR is responsible for the generation of subgenomic flavivirus RNA (sfRNA) that helps the virus evade host immune response, thereby affecting viral pathogenesis and the development of vaccines.
Journal ArticleDOI
Structure mapping of dengue and Zika viruses reveals functional long-range interactions
Roland G. Huber,Xin Ni Lim,Wy Ching Ng,Adelene Y. L. Sim,Hui Xian Poh,Yang Shen,Su Ying Lim,Anna Karin Beatrice Sundstrom,Xuyang Sun,Jong Ghut Aw,Horng Khit Too,Peng Hee Boey,Andreas Wilm,Tanu Chawla,Milly M. Choy,Milly M. Choy,Lu Jiang,Lu Jiang,Paola Florez de Sessions,Xian Jun Loh,Xian Jun Loh,Sylvie Alonso,Martin L. Hibberd,Martin L. Hibberd,Niranjan Nagarajan,Eng Eong Ooi,Peter J. Bond,October M. Sessions,Yue Wan,Yue Wan,Yue Wan +30 more
TL;DR: Detailed analyses of viral RNA structure in virions and in infected cells for four dengue virus serotypes and four Zika virus strains are provided, and conserved structures that are important for virus replication are identified.
Journal ArticleDOI
A systematic approach to the development of a safe live attenuated Zika vaccine
Swee Sen Kwek,Satoru Watanabe,Kuan Rong Chan,Eugenia Z. Ong,Hwee Cheng Tan,Wy Ching Ng,Mien T. X. Nguyen,Esther S. Gan,Summer L. Zhang,Kitti Wing Ki Chan,Jun Hao Tan,October M. Sessions,Menchie Manuel,Julien Pompon,Julien Pompon,Camillus Chua,Sharifah Nur Hazirah,Karl Tryggvason,Karl Tryggvason,Subhash G. Vasudevan,Eng Eong Ooi +20 more
TL;DR: A ZIKV variant that produces small plaques due to interferon (IFN)-restricted viral propagation and displays attenuated infection of endothelial cells is identified and it is shown that these properties collectively reduce the risk of organ infections and vertical transmission in a mouse model but remain sufficiently immunogenic to prevent wild-type ZikV infection.
Journal ArticleDOI
Identification and characterization of host proteins bound to dengue virus 3' UTR reveal an antiviral role for quaking proteins.
Kuo-Chieh Liao,Vanessa Chuo,Wy Ching Ng,Suat Peng Neo,Julien Pompon,Julien Pompon,Jayantha Gunaratne,Jayantha Gunaratne,Eng Eong Ooi,Eng Eong Ooi,Mariano A. Garcia-Blanco +10 more
TL;DR: A novel function of QKI is described in restricting viral replication by mapping the QKI interaction site and identifying a QKI response element in DENV4 3' UTR.
Journal ArticleDOI
A Non-structural 1 Protein G53D Substitution Attenuates a Clinically Tested Live Dengue Vaccine.
Milly M. Choy,Dorothy H.L. Ng,Dorothy H.L. Ng,Tanamas Siriphanitchakorn,Wy Ching Ng,Karin B. Sundstrom,Hwee Cheng Tan,Summer L. Zhang,Kitti Wing Ki Chan,Menchie Manuel,R. Manjunatha Kini,Kuan Rong Chan,Subhash G. Vasudevan,Eng Eong Ooi +13 more
TL;DR: The site-directed mutagenesis on a 16681 infectious clone identifies a single G53D substitution in the non-structural 1 (NS1) protein that reduces 16681 infection and dissemination in both Aedes aegypti, as well as in mammalian cells to produce the characteristic phenotypes of PDK53.
