TL;DR: This work integrates multiple transcript collections to generate a comprehensive atlas of 27,919 human lncRNA genes with high-confidence 5′ ends and expression profiles across 1,829 samples from the major human primary cell types and tissues, identifying 19,175 potentially functional lncRNAs in the human genome.
Journal ArticleDOI
BCL11A enhancer dissection by Cas9-mediated in situ saturating mutagenesis
Matthew C. Canver,Elenoe C. Smith,Falak Sher,Luca Pinello,Neville E. Sanjana,Ophir Shalem,Diane D. Chen,Patrick G. Schupp,Divya S. Vinjamur,Sara P. Garcia,Sidinh Luc,Ryo Kurita,Yukio Nakamura,Yuko Fujiwara,Yuko Fujiwara,Takahiro Maeda,Guo-Cheng Yuan,Feng Zhang,Stuart H. Orkin,Stuart H. Orkin,Daniel E. Bauer +20 more
TL;DR: A pooled clustered regularly interspaced palindromic repeat-Cas9 guide RNA libraries are developed to perform in situ saturating mutagenesis of the human and mouse enhancers and reveal critical minimal features and discrete vulnerabilities of these enhancers.
Journal ArticleDOI
Establishment of immortalized human erythroid progenitor cell lines able to produce enucleated red blood cells.
Ryo Kurita,Noriko Suda,Kazuhiro Sudo,Kenichi Miharada,Takashi Hiroyama,Hiroyuki Miyoshi,Kenzaburo Tani,Yukio Nakamura +7 more
TL;DR: This is the first demonstration of the feasibility of using immortalized human erythroid progenitor cell lines as an ex vivo source for production of enucleated RBCs, and these immortalized cell lines all produce enucleations after induction of differentiation in vitro.
Journal ArticleDOI
A genome-editing strategy to treat β-hemoglobinopathies that recapitulates a mutation associated with a benign genetic condition
Elizabeth A. Traxler,Elizabeth A. Traxler,Yu Yao,Yong Dong Wang,Kaitly J. Woodard,Ryo Kurita,Yukio Nakamura,Jim R. Hughes,Ross C. Hardison,Gerd A. Blobel,Chunliang Li,Mitchell J. Weiss +11 more
TL;DR: Here, CRISPR–Cas9-mediated genome editing of human blood progenitors to mutate a 13-nt sequence that is present in the promoters of the HBG1 and HBG2 genes, thereby recapitulating a naturally occurring HPFH-associated mutation are performed.
Journal ArticleDOI
Transcription factors LRF and BCL11A independently repress expression of fetal hemoglobin
Takeshi Masuda,Xin Wang,Manami Maeda,Matthew C. Canver,Falak Sher,Alister P. W. Funnell,Chris Fisher,Maria C. Suciu,Gabriella E. Martyn,Laura J. Norton,Catherine Zhu,Ryo Kurita,Yukio Nakamura,Jian Xu,Douglas R. Higgs,Merlin Crossley,Daniel E. Bauer,Stuart H. Orkin,Peter V. Kharchenko,Takahiro Maeda +19 more
TL;DR: It is found that the LRF/ZBTB7A transcription factor occupies fetal γ-globin genes and maintains the nucleosome density necessary for γ -globin gene silencing in adults, and that LRF confers its repressive activity through a NuRD repressor complex independent of the fetal globin repressor BCL11A.
