Abstract: Understanding gene expression patterns in response to altered environmental conditions at different time points of the preimplantation period would improve our knowledge on regulation of embryonic development. Here we aimed to examine the effect of alternative in vivo and in vitro culture conditions at the time of major embryonic genome activation (EGA) on the development and transcriptome profile of bovine blastocysts. Four different blastocyst groups were produced under alternative in vivo and in vitro culture conditions before or after major EGA. Completely in vitro- and in vivo-produced blastocysts were used as controls. We compared gene expression patterns between each blastocyst group and in vivo blastocyst control group using EmbryoGENE's bovine microarray. The data showed that changing culture conditions from in vivo to in vitro or vice versa, either before or after the time of major EGA, had no effect on the developmental rates; however, in vitro conditions during that time critically influenced the transcriptome of the blastocysts produced. The source of oocyte had a critical effect on developmental rates and the ability of the embryo to react to changing culture conditions. Ontological classification highlighted a marked contrast in expression patterns for lipid metabolism and oxidative stress response between blastocysts generated in vivo versus in vitro, with opposite trends. Molecular mechanisms and pathways that are influenced by altered culture conditions during EGA were defined. These results will help in the development of new strategies to modify culture conditions at this critical stage to enhance the development of competent blastocysts.

Abstract: Hepatitis C virus (HCV) infection is a major public health burden in Egypt, where it bears the highest prevalence rate in the world Estimates for prevalence are based upon data reported from the 2008 and 2015 Egypt Demographic Health Surveys In this review, we demonstrate the prevalence results of both surveys and analyze the difference in the results The overall HCV prevalence is estimated to be declining However, the clinical impact of chronic HCV infection is expected to grow considerably A mathematical model shows that by increasing the rate of treatment, the expected number of patients will decline significantly in 2030 The current and expected future burden of chronic HCV infection to the Egyptian economy, including direct and indirect costs due to disability and loss of lives, has been estimated and discussed in this review The economic burden will continue to grow, but a model shows that the introduction of highly effective therapies will result in a significant reduction in the cumulative total economic burden of HCV by 2030 In recognition of the HCV tremendous health and economic burden, the Egyptian government established the National Committee for Control of Viral Hepatitis to implement an integrated nationwide strategy to provide patient care and ensure global treatment access This review illustrates the epidemiological and disease burden aspects of HCV in Egypt in addition to introducing the national plan and program for managing HCV, which has been successful so far in treating a large number of patients, with the aim of achieving disease control and eventual elimination in Egypt

Abstract: Background The present study was conducted to investigate the chemopreventive effects of garlic extract and silymarin on N-nitrosodiethylamine (NDEA) and carbon tetrachloride (CCl(4))-induced hepatotoxicity in male albino rats. Methods and results Animals were pretreated with garlic, silymarin or both for one week prior to the injection of NDEA. Then animals received a single injection of NDEA followed by weekly subcutaneous injections of CCl(4) for 6 weeks. Oral administration was then continued along with the injection of CCl(4) for the duration of the experiment. Serum aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), hepatic lipid peroxidation (LPO), superoxide dismutase (SOD), reduced glutathione (GSH), glutathione-S-transferase (GST) and glutathione reductase (GSR) were measured. Injection of NDEA induced a significant elevation in serum AST, ALT and ALP. In the liver, NDEA increased oxidative stress through the increase in LPO and decrease in SOD, and GSH-dependent enzymes. Although administration of garlic or silymarin significantly reduced the liver toxicity, combined administration was more effective in preventing the development of hepatotoxicity. Conclusion These novel findings suggest that silymarin and garlic have a synergistic effect, and could be used as hepatoprotective agents against hepatotoxicity.

Abstract: In this paper, the problem of simultaneous scheduling of machines and identical automated guided vehicles (AGVs) in flexible manufacturing systems is addressed with the objective of minimizing the makespan. This problem is composed of two interrelated decision problems: the scheduling of machines, and the scheduling of AGVs. Both problems are known to be NP-complete, resulting in a more complicated NP-complete problem when they are considered simultaneously. A new hybrid Genetic-algorithm/heuristic coding scheme is developed for the studied problem. The developed coding scheme is combined with a set of genetic algorithm (GA) operators selected from the literature of the applications of GAs to the scheduling problems. The algorithm is applied to a set of 82 test problems, which was constructed by other researchers, and the comparison of the results indicates the superior performance of the developed coding.

Abstract: The cutting fluid is significant in any metal cutting operation, for cooling the cutting tool and the surface of the workpiece, by lubricating the tool-workpiece interface and removing chips from the cutting zone. Recently, many researchers have been focusing on minimum quantity lubrication (MQL) among the numerous methods existing on the application of the coolant as it reduces the usage of coolant by spurting a mixture of compressed air and cutting fluid in an improved way instead of flood cooling. The MQL method has been demonstrated to be appropriate as it fulfills the necessities of ‘green’ machining. In the current study, firstly, various lubrication methods were introduced which are used in machining processes, and then, basic machining processes used in manufacturing industries such as grinding, milling, turning, and drilling have been discussed. The comprehensive review of various nanofluids (NFs) used as lubricants by different researchers for machining process is presented. Furthermore, some cases of utilizing NFs in machining operations have been reported briefly in a table. Based on the studies, it can be concluded that utilizing NFs as coolant and lubricant lead to lower tool temperature, tool wear, higher surface quality, and less environmental dangers. However, the high cost of nanoparticles, need for devices, clustering, and sediment are still challenges for the NF applications in metalworking operations. At last, the article identifies the opportunities for using NFs as lubricants in the future. It should be stated that this work offers a clear guideline for utilizing MQL and MQL-nanofluid approaches in machining processes. This guideline shows the physical, tribological, and heat transfer mechanisms associated with employing such cooling/lubrication approaches and their effects on different machining quality characteristics such as tool wear, surface integrity, and cutting forces.
