Abstract: High-fat (HF) diet modulates gut microbiota and increases plasma concentration of lipopolysaccharide (LPS) which is associated with obesity and its related low-grade inflammation and cognitive decline. Rhein is the main ingredient of the rhubarb plant which has been used as an anti-inflammatory agent for several millennia. However, the potential effects of rhein against HF diet-induced obesity and its associated alteration of gut microbiota, inflammation and cognitive decline have not been studied. In this study, C57BL/6J male mice were fed an HF diet for 8 weeks to induce obesity, and then treated with oral rhein (120 mg/kg body weight/day in HF diet) for a further 6 weeks. Chronic rhein treatment prevented the HF diet-induced recognition memory impairment assessed by the novel object recognition test, neuroinflammation and brain-derived neurotrophic factor (BDNF) deficits in the perirhinal cortex. Furthermore, rhein inhibited the HF diet-induced increased plasma LPS level and the proinflammatory macrophage accumulation in the colon and alteration of microbiota, including decreasing Bacteroides – Prevotella spp. and Desulfovibrios spp. DNA and increasing Bifidobacterium spp. and Lactobacillus spp. DNA. Moreover, rhein also reduced body weight and improved glucose tolerance in HF diet-induced obese mice. In conclusion, rhein improved recognition memory and prevented obesity in mice on a chronic HF diet. These beneficial effects occur via the modulation of microbiota, hypoendotoxinemia, inhibition of macrophage accumulation, anti-neuroinflammation and the improvement of BDNF expression. Therefore, supplementation with rhein-enriched food or herbal medicine could be beneficial as a preventive strategy for chronic HF diet-induced cognitive decline, microbiota alteration and neuroinflammation.

Abstract: Tumor treatment is still complicated in the field of medicine. Tumor immunotherapy has been the most interesting research field in cancer therapy. Application of chimeric antigen receptor T (CAR-T) cell therapy has recently achieved excellent clinical outcome in patients, especially those with CD19-positive hematologic malignancies. This phenomenon has induced intense interest to develop CAR-T cell therapy for cancer, especially for solid tumors. However, the performance of CAR-T cell treatment in solid tumor is not as satisfactory as that in hematologic disease. Clinical studies on some neoplasms, such as glioblastoma, ovarian cancer, and cholangiocarcinoma, have achieved desirable outcome. This review describes the history and evolution of CAR-T, generalizes the structure and preparation of CAR-T, and summarizes the latest advances on CAR-T cell therapy in different tumor types. The last section presents the current challenges and prospects of CAR-T application to provide guidance for subsequent research.

Abstract: Information coding strategies are becoming increasingly crucial due to the storage demand brought by the information explosion. In particular, bioinformation coding has attracted great attention for its advantages of excellent storage capacity and long lifetime. Herein, we present an innovative bioinspired MXene-integrated photonic crystal (PhC) array for multichannel bioinformation coding. PhC arrays with similar structure to Stenocara beetle's back are utilized as the substrate, exhibiting properties of high throughput and stability. MXene nanosheets are further integrated on the PhC array's substrate with the assistance of the adhesion capacity of mussel-inspired dopamine (DA). Benefitting from their fluorescence resonance energy transfer effect, MXene nanosheets can quench the fluorescence signals of quantum dot (QD) modified DNA probes unless the corresponding targets exist. Additionally, these black MXene nanosheets can enhance the contrast of structural color. In this case, the encrypted information can be easily read out by simply observing the fluorescence signal of DNA probes. It is demonstrated that this strategy based on bioinspired MXene-integrated PhC arrays can realize high-throughput information encoding and encryption, which opens a chapter of bioinformation coding.
