TL;DR: Overall survival was significantly longer in the pembrolizumab group than in the chemotherapy group in all three TPS populations, and the benefit-to-risk profile suggests that first-line pembrology monotherapy can be extended as first line therapy for locally advanced or metastatic non-small-cell lung cancer patients with sensitising EGFR or ALK translocation.

TL;DR: As compared with crizotinib, alectinib showed superior efficacy and lower toxicity in primary treatment of ALK‐positive NSCLC and independent review committee–assessed progression‐free survival were consistent with those for the primary end point.

Non–Small Cell Lung Cancer: Epidemiology, Screening, Diagnosis, and Treatment
TL;DR: The results of new trials continue to help us understand the role of these novel agents and which patients are more likely to benefit; ICIs are now part of the first-line NSCLC treatment armamentarium as monotherapy, combined with chemotherapy, or after definite chemoradiotherapy in patients with stage III unresectable NSCLCs.

Identification of the transforming EML4–ALK fusion gene in non-small-cell lung cancer
Manabu Soda,Young Lim Choi,Munehiro Enomoto,Shuji Takada,Yoshihiro Yamashita,Shunpei Ishikawa,Shin-ichiro Fujiwara,Hideki Watanabe,Kentaro Kurashina,Hisashi Hatanaka,Masashi Bando,Shoji Ohno,Yuichi Ishikawa,Hiroyuki Aburatani,Toshiro Niki,Yasunori Sohara,Yukihiko Sugiyama,Hiroyuki Mano +17 more
TL;DR: It is shown that a small inversion within chromosome 2p results in the formation of a fusion gene comprising portions of the echinoderm microtubule-associated protein-like 4 (EML4) gene and the anaplastic lymphoma kinase (ALK) gene in non-small-cell lung cancer (NSCLC) cells.

Lee,Anders Mellemgaard,Keunchil Park,Shehkar Patil,Janusz Rolski,Tuncay Göksel,Filippo de Marinis,Lorinda Simms,Katherine P. Sugarman,David R. Gandara +19 more
TL;DR: This is the first prospective phase III study in NSCLC to show survival differences based on histologic type and cisplatin/pemetrexed provides similar efficacy with better tolerability and more convenient administration than cis platin/gemcitabine.

First-line crizotinib versus chemotherapy in ALK-positive lung cancer
Benjamin Solomon,Tony Mok,Dong Wan Kim,Yi-Long Wu,Kazuhiko Nakagawa,Tarek Mekhail,Enriqueta Felip,Federico Cappuzzo,Jolanda Paolini,Tiziana Usari,Shrividya Iyer,Arlene Reisman,Keith D. Wilner,Jennifer M. Tursi,Fiona H Blackhall +14 more
TL;DR: Crizotinib was superior to standard first-line pemetrexed-plus-platinum chemotherapy in patients with previously untreated advanced ALK-positive NSCLC and was associated with greater reduction in lung cancer symptoms and greater improvement in quality of life.

Clinical Features and Outcome of Patients With Non–Small-Cell Lung Cancer Who Harbor EML4-ALK
Alice T. Shaw,Beow Y. Yeap,Mari Mino-Kenudson,Subba R. Digumarthy,Daniel B. Costa,Rebecca S. Heist,Benjamin Solomon,Hannah Stubbs,Sonal Admane,Ultan McDermott,Jeffrey Settleman,Susumu Kobayashi,Eugene J. Mark,Scott J. Rodig,Lucian R. Chirieac,Eunice L. Kwak,Thomas J. Lynch,A. John Iafrate +17 more
TL;DR: EML4-ALK defines a molecular subset of NSCLC with distinct clinical characteristics and patients who harbor this mutation do not benefit from EGFR TKIs and should be directed to trials of ALK-targeted agents.
