Abstract: Cell suspension cultures of soybean (Glycine max L.) were incubated with [¹⁴C]-2,4- dichlorophenoxyacetic acid (62 h, 27 °C). The cells were converted to protoplasts by use of the cell-wall degrading enzymes, Driselase and pectinase. This procedure resulted in the release of the cellular amino acid conjugates as well as part of the free 2,4-dichlorophenoxyacetic acid and of the β-ᴅ- glucoside conjugates. A vacuole fraction was obtained by mild osmotic shock in 10% yield, and was characterized by its polypeptide composition. The purified vacuoles were enriched in β-ᴅ-glucoside conjugates, free 2,4-dichlorophenoxyacetic acid being only a minor vacuolar constituent, and amino acid conjugates not being detectable.

Abstract: A new xanthone glucoside named isoswertianolin (2) was isolated from Swertia japonica and its structure was elucidated to be 5-O-β-glucopyranoside of bellidifolin by 1H and 13C nuclear magnetic resonance (NMR) spectroscopy. In connection with this study, the structure of swertianolin (1), a constituent of this plant, was reinvestigated by NMR spectroscopy, leading to the revision of its structure (the location of its glucosyl linkage) from 1-O-β-glucopyranoside of bellidifolin (3) to 8-O-β-glucopyranoside of 3. It follows that 1 is identical with the glucoside isolated from Gentiana campestris by Kaldas et al. in 1974. Analogously, the structure of norswertianolin (6), previously isolated from Swertia randaiensis HAYATA, was also revised, being formulated as 8-O-β-glucopyranoside of desmethylbellidifolin (7).

Abstract: A spontaneous mutant of Streptococcus salivarius ATCC 25975 was isolated by inoculating an agar medium containing 11 mM lactose and 0.5 mM 2-deoxyglucose. This mutant grew poorly on 5 mM glucose but almost as well as the parental strain on 110 mM glucose. Uptake of 2-deoxyglucose was abolished by the mutation, and phosphoenolpyruvate: glucose phosphotransferase activity could not be detected with toluenized cells under normal conditions when the glucose concentration was below 5 mM. Data from growth experiments, glycolysis, and uptake studies indicated the presence of a second phosphoenolpyruvate: glucose phosphotransferase system that could catalyze the phosphorylation of alpha-methyl glucoside. The activity of this system was detected by a spectrophotometric assay coupled with lactate dehydrogenase and by a radioactive isotope method using methyl alpha-D-[U-14C] glucoside. The phosphorylation was phosphoenolpyruvate dependent. The apparent Km of the system for glucose and alpha-methyl glucoside was approximately 20 mM. Studies with energy poisons ruled out the possibility of an active transport system, and accumulation of alpha-methyl glucoside argued against facilitated diffusion. It was concluded that the other glucose transport system which allowed growth of the mutant strain of S. salivarius was a second phosphoenolpyruvate: glucose phosphotransferase system.

Abstract: (±)-[2-14C]abscisic acid (ABA) and [2-14C]dihydrophaseic acid (DPA) fed to tomato shoots were converted into an acidic, polar conjugate that was hydrolysed (1 M HCl, 100°C) to D-glucose and DPA. The tetraacetylmethyl ester of the metabolite was isolated by high performance liquid chromatography as colourless crystals (m.p. 81-84°). N.m.r. spectra showed the presence of a β-glucosidic linkage (J 7 Hz, CDCl3; 8 Hz, D5 pyridine) between C-4' of DPA and C-1 of glucose. Mass spectra of the 0-acetylmethyl ester of the conjugate, and also of material biosynthesized from [1'-18O]ABA, showed that the glucoside is at C-4' of DPA. The abbreviation 'DPAGS' is proposed.

Abstract: A restrained, whole animal preparation was used to study chemical sensitivity and specificity of three cells in the medial galeal chemosensillum of adult red turnip beetles. One cell is sensitive to sugars, particularly sucrose and maltose, two cells respond to glucosinolates and two to the glucoside arbutin. Responses to mixtures of these compounds revealed that arbutin stimulates the sugar-sensitive cell and one of the glucosinolate-sensitive cells. The second glucosinolate-sensitive cell was stimulated only by glucosinolates. Dose-response curves were determined for most of the compounds tested.

Abstract: Acarbose was administered at a dose of 300 mg/day fbr S weeks to nine healthy volunteers (Table 1). Volunteers were doctors, nurses and laboratory technicians in Keio University Hospital. All were requested not to change their dietary habits and not to drink much alcohol. Blood was collected serially after 12 hours of fasting. Very low density lipoprotein (VLDL) was separated ultracentrifugally by the method of Havel et aL (5). High density lipoprotein (HDL) was separated by the heparin-manganese precipitation method (16). Abstract: Adiposin-1 and -2 exhibited potent inhibitory activities against alpha-amylase, human salivary alpha-amylase and disaccharidases isolated from porcine small intestine. The effect of adiposin-1 and -2 on hydrolysis of glucoamylase was non-competitive. Adiposin showed antimicrobial activities against some Gram-positive bacteria, Gram-negative bacteria belonging to Enterobacteriaceae, Some anaerobic bacteria and some phytopathogenic fungi, and showed a synergistic effect on the antibacterial activity with some maltooligosaccharides. However, these antibacterial activities were suppressed by addition of various other saccharides.

Abstract: Several anthraglycosides were isolated from the bark of Rhamnus alaternus L. These had the following Rf values when chromatographed on Whatman no. 1 paper in the solvent system n-propanol, ethyl acetate, water (40:30:30 v/v): G1, 0.98; G2, 0.92; G3, -; G4, 0.79; G5, 0.72; G6, 0.65; G7, 0.61. However, in the TLC system chloroform, ethanol-95%, water (60:30:2 v/v) on silica gel G, G5 was found to be a mixture of G5a and G5b, the latter with the lower Rf value; while G7 separated into G7a, G7b and G7c in descending order of their Rf values. G2, highest in yield, is an emodin glucoside; G4 is an alaternin glucoside; G1 occurs in least amount. G1, G5b, and G6 are diglycosides of emodin, each yielding glucose and rhamnose. G6 hydrolyzes into G2 on TLC in the solvent system methanol, ethyl acetate, water (17:100:13 v/v) on silica gel G. G5a yields physcion, glucose and rhamnose.
