Riggins,Rachel Karchin,Giovanni Parmigiani,Darell D. Bigner,Hai Yan,Nick Papadopoulos,Bert Vogelstein,Kenneth W. Kinzler,Victor E. Velculescu +49 more
TL;DR: Key differences between the genetic landscapes of adult and childhood cancers are demonstrated, dysregulation of developmental pathways as an important mechanism underlying MBs is highlighted, and a role for a specific type of histone methylation in human tumorigenesis is identified.
Journal ArticleDOI
Akt-Dependent Metabolic Reprogramming Regulates Tumor Cell Histone Acetylation
Joyce V. Lee,Alessandro Carrer,Supriya Shah,Nathaniel W. Snyder,Shuanzeng Wei,Sriram Venneti,Andrew J. Worth,Zuo-Fei Yuan,Hee-Woong Lim,Shichong Liu,Ellen Jackson,Nicole M. Aiello,Naomi B. Haas,Timothy R. Rebbeck,Alexander R. Judkins,Kyoung-Jae Won,Lewis A. Chodosh,Benjamin A. Garcia,Ben Z. Stanger,Michael Feldman,Ian A. Blair,Kathryn E. Wellen +21 more
TL;DR: It is demonstrated that acetyl-CoA is dynamically regulated by glucose availability in cancer cells and that the ratio of acetyl -CoA:coenzyme A within the nucleus modulates global histone acetylation levels.
Journal ArticleDOI
Hypoxia Induces Production of L-2-Hydroxyglutarate
Andrew M. Intlekofer,Raymond G. DeMatteo,Sriram Venneti,Lydia W.S. Finley,Chao Lu,Alexander R. Judkins,Ariën S. Rustenburg,Patrick B. Grinaway,John D. Chodera,Justin R. Cross,Craig B. Thompson +10 more
TL;DR: It is reported that under conditions of oxygen limitation, mammalian cells selectively produce L-2HG via enzymatic reduction of α-KG through promiscuous substrate usage primarily by lactate dehydrogenase A (LDHA).
Journal ArticleDOI
Immunohistochemical analysis of hSNF5/INI1 in pediatric CNS neoplasms.
TL;DR: Infection with an antibody to INI1 may have particular utility in the analysis of tumors with indeterminate histologic features or atypical immunophenotypic profiles, and may be useful in confirming the histologic diagnosis.
Journal ArticleDOI
Asparagine plays a critical role in regulating cellular adaptation to glutamine depletion
Ji Zhang,Jing Fan,Sriram Venneti,Justin R. Cross,Toshimitsu Takagi,Bhavneet Bhinder,Hakim Djaballah,Masayuki Kanai,Emily H. Cheng,Alexander R. Judkins,Bruce R. Pawel,Julie E. Baggs,Sara Cherry,Joshua D. Rabinowitz,Craig B. Thompson +14 more
TL;DR: It was found that asparagine was necessary and sufficient to suppress glutamine-withdrawal-induced apoptosis without restoring the levels of other nonessential amino acids or TCA cycle intermediates.
