Journal ArticleDOI
Stromal elements act to restrain, rather than support, pancreatic ductal adenocarcinoma
Andrew D. Rhim,Andrew D. Rhim,Paul E. Oberstein,Dafydd H. Thomas,Emily T. Mirek,Carmine F. Palermo,Stephen A. Sastra,Erin N. Dekleva,Tyler Saunders,Claudia P. Becerra,Ian W. Tattersall,C. Benedikt Westphalen,Jan Kitajewski,Maite G. Fernandez-Barrena,Martin E. Fernandez-Zapico,Christine A. Iacobuzio-Donahue,Kenneth P. Olive,Ben Z. Stanger +17 more
TL;DR: De deleting Shh in a well-defined mouse model of PDAC demonstrated that some components of the tumor stroma can act to restrain tumor growth, and administration of VEGFR blocking antibody selectively improved survival of Shh-deficient tumors.
Journal ArticleDOI
Cysteine depletion induces pancreatic tumor ferroptosis in mice.
Michael A. Badgley,Daniel M. Kremer,H. Carlo Maurer,H. Carlo Maurer,Kathleen E. DelGiorno,Ho-Joon Lee,Vinee Purohit,Irina Sagalovskiy,Alice Ma,Jonathan Kapilian,Christina E. M. Firl,Amanda R. Decker,Steve A. Sastra,Carmine F. Palermo,Leonardo R. Andrade,Peter Sajjakulnukit,Li Zhang,Zachary P. Tolstyka,Tal Hirschhorn,Candice Lamb,Tong Liu,Wei Gu,E. Scott Seeley,Everett Stone,George Georgiou,Uri Manor,Alina Iuga,Geoffrey M. Wahl,Brent R. Stockwell,Costas A. Lyssiotis,Kenneth P. Olive +30 more
TL;DR: This work shows that the import of oxidized cysteine (cystine) via system xC– is a critical dependency of pancreatic ductal adenocarcinoma (PDAC), which is a leading cause of cancer mortality.
Journal ArticleDOI
Preclinical Pharmacologic Evaluation of Pralatrexate and Romidepsin Confirms Potent Synergy of the Combination in a Murine Model of Human T-cell Lymphoma
Salvia Jain,Xavier Jirau-Serrano,Kelly Zullo,Luigi Scotto,Carmine F. Palermo,Stephen A. Sastra,Kenneth P. Olive,Serge Cremers,Tiffany Thomas,Ying Wei,Yuan Zhang,Govind Bhagat,Jennifer E Amengual,Changchun Deng,Charles Karan,Ronald Realubit,Susan E. Bates,Owen A. O'Connor +17 more
TL;DR: It is suggested that the combination of pralatrexate and romidepsin merits clinical study in patients with TCLs using complementary imaging modalities, such as SBL and 3D-US.
Journal ArticleDOI
Noninvasive Young's modulus visualization of fibrosis progression and delineation of pancreatic ductal adenocarcinoma (PDAC) tumors using Harmonic Motion Elastography (HME) in vivo.
Alireza Nabavizadeh,Thomas Payen,Alina Iuga,Irina Sagalovskiy,Deborah D. Desrouilleres,Niloufar Saharkhiz,Carmine F. Palermo,Stephen A. Sastra,Paul E. Oberstein,Vilma L. Rosario,Michael D. Kluger,Beth Schrope,John A. Chabot,Kenneth P. Olive,Elisa E. Konofagou +14 more
TL;DR: HME, a quantitative ultrasound-based imaging method, was applied to calculate Young's modulus (YM) in PDAC mouse models and human pancreatic resection specimens of PDAC to develop a non-invasive, quantitative imaging tool for detecting, staging and delineating PDAC tumor margins based on the change in collagen density.
Journal ArticleDOI
Harmonic motion imaging for abdominal tumor detection and high-intensity focused ultrasound ablation monitoring: an in vivo feasibility study in a transgenic mouse model of pancreatic cancer
Hong Chen,Gary Y. Hou,Yang Han,Thomas Payen,Carmine F. Palermo,Kenneth P. Olive,Elisa E. Konofagou +6 more
TL;DR: The feasibility of HMI in abdominal tumor detection and HIFU ablation monitoring was demonstrated and no tissue damage was associated with HMI when it was used for the sole purpose of elasticity imaging.
