Surfen, a small molecule antagonist of heparan sulfate
Manuela Schuksz,Mark M. Fuster,Jillian R. Brown,Brett E. Crawford,David Ditto,Roger Lawrence,Charles A. Glass,Lianchun Wang,Yitzhak Tor,Jeffrey D. Esko +9 more
TL;DR: The findings demonstrate the feasibility of identifying small molecule antagonists of heparan sulfate and raise the possibility of developing pharmacological agents to treat disorders that involve glycosaminoglycan–protein interactions.
Journal ArticleDOI
Metabolic engineering of Chinese hamster ovary cells: Towards a bioengineered heparin
Jong Youn Baik,Leyla Gasimli,Bo Yang,Payel Datta,Fuming Zhang,Charles A. Glass,Jeffrey D. Esko,Robert J. Linhardt,Susan T. Sharfstein +8 more
TL;DR: Disaccharide analysis of the engineered HS showed a substantial increase in N-sulfo groups, but did not show a pattern consistent with pharmacological heparin, suggesting that further balancing the expression of transgenes with the expression levels of endogenous enzymes involved in HS/heparin biosynthesis might be necessary.
Journal ArticleDOI
A disaccharide-based inhibitor of glycosylation attenuates metastatic tumor cell dissemination.
TL;DR: Data show that AcGnG-NM provides a targeted glycoside-based therapy for the treatment of hematogenous dissemination of tumor cells and on spontaneous hematogenously borne malignant cells facilitates metastasis.

SARS-CoV-2 Infection Depends on Cellular Heparan Sulfate and ACE2
Thomas Mandel Clausen,Thomas Mandel Clausen,Daniel R. Sandoval,Charlotte B. Spliid,Charlotte B. Spliid,Jessica Pihl,Jessica Pihl,Chelsea D. Painter,Chelsea D. Painter,Bryan E. Thacker,Charles A. Glass,Anoop Narayanan,Sydney A. Majowicz,Yang Zhang,Jonathan L. Torres,Gregory J. Golden,Gregory J. Golden,Ryan Porell,Aaron F. Garretson,Logan K. Laubach,Jared Feldman,Xin Yin,Yuan Pu,Blake M. Schmidt,Kamil Godula,Joyce Jose,Kevin D. Corbett,Andrew B. Ward,Aaron F. Carlin,Jeffrey D. Esko +39 more
TL;DR: It is shown that SARS-CoV-2 spike protein interacts with cell surface heparan sulfate and angiotensin converting enzyme 2 (ACE2) through its Receptor Binding Domain, and docking studies suggest a putative heparin/heparin sulfate-binding site adjacent to the domain that binds to ACE2.
Journal ArticleDOI
Bioengineering murine mastocytoma cells to produce anticoagulant heparin.
Leyla Gasimli,Charles A. Glass,Payel Datta,Bo Yang,Guoyun Li,Trent R. Gemmill,Jong Youn Baik,Susan T. Sharfstein,Jeffrey D. Esko,Robert J. Linhardt,Robert J. Linhardt +10 more
TL;DR: It is shown that transfection of MST cells with a retroviral vector containing heparan sulfate 3-O-sulfotransferase-1 (Hs3st1) restores anticoagulant activity and contributes toward a better understanding of the HP biosynthetic pathway.
