Cholesterol Oxidation Products Are Sensitive and Specific Blood-Based Biomarkers for Niemann-Pick C1 Disease
Forbes D. Porter,David E. Scherrer,Michael H. Lanier,S. Joshua Langmade,Vasumathi Molugu,Sarah E. Gale,Dana Olzeski,Rohini Sidhu,Dennis J. Dietzen,Rao Fu,Christopher A. Wassif,Nicole M. Yanjanin,Steven P. Marso,John A. House,Charles H. Vite,Jean E. Schaffer,Daniel S. Ory +16 more
TL;DR: Blood concentrations of two related oxysterols molecules were almost 10 times higher in Niemann-Pick C1 patients than in age-matched healthy controls or those with other diseases such as atherosclerosis or diabetes, suggesting that the two oxysterol molecules are accurate diagnostic markers of early clinical disease and can be used not only to monitor disease progression but also to demonstrate drug efficacy.
Journal ArticleDOI
Intrathecal 2-hydroxypropyl-β-cyclodextrin decreases neurological disease progression in Niemann-Pick disease, type C1: a non-randomised, open-label, phase 1–2 trial
Daniel S. Ory,Elizabeth A. Ottinger,Nicole Y. Farhat,Kelly A. King,Xuntian Jiang,Lisa Weissfeld,Elizabeth Berry-Kravis,Cristin Davidson,Simona Bianconi,Lee Ann Keener,Ravichandran Rao,Ariane Soldatos,Rohini Sidhu,Kimberly A Walters,Xin Xu,Audrey Thurm,Beth Solomon,William J. Pavan,Bernardus Nicolaas Machielse,Mark L. Kao,Steven A. Silber,John C. McKew,Carmen C. Brewer,Charles H. Vite,Steven U. Walkley,Christopher P. Austin,Forbes D. Porter +26 more
TL;DR: This open-label, dose-escalation phase 1-2a study gave monthly intrathecal HPβCD to participants with NPC1 with neurological manifestation at the National Institutes of Health and evaluated Serum and CSF 24(S)-hydroxycholesterol (24[S]-HC), which serves as a biomarker of target engagement, andCSF protein biomarkers were evaluated.
Journal ArticleDOI
Feline intracranial neoplasia: Retrospective review of 160 cases (1985-2001)
Mark T. Troxel,Charles H. Vite,Thomas J. Van Winkle,Alisa L. Newton,Deena Tiches,Betsy Dayrell-Hart,Amy S. Kapatkin,Frances S. Shofer,Sheldon A. Steinberg +8 more
TL;DR: It was found that lesion location was predictive of tumor type with diffuse cerebral or brainstem involvement predictive of lymphoma and third ventricle involved predictive of meningioma.
Journal ArticleDOI
Crowdsourcing reproducible seizure forecasting in human and canine epilepsy
Benjamin H. Brinkmann,Joost B. Wagenaar,Drew Abbot,Phillip Adkins,Simone C. Bosshard,Min Chen,Quang M. Tieng,Jialune He,F. J. Muñoz-Almaraz,Paloma Botella-Rocamora,Juan Pardo,Francisco Zamora-Martínez,Michael Hills,Wei Wu,Iryna Korshunova,William Cukierski,Charles H. Vite,Edward E. Patterson,Brian Litt,Gregory A. Worrell +19 more
TL;DR: An online, open-access seizure forecasting competition using intracranial EEG recordings from canines with naturally occurring epilepsy and human patients undergoing presurgical monitoring wins, with the winning algorithms forecast seizures at rates significantly greater than chance.
Journal ArticleDOI
Intracisternal cyclodextrin prevents cerebellar dysfunction and Purkinje cell death in feline Niemann-Pick type C1 disease
Charles H. Vite,Jessica H. Bagel,Gary P. Swain,Maria Prociuk,Tracey Sikora,Veronika M. Stein,Patricia O'Donnell,Therese Ruane,Sarah Ward,Alexandra Crooks,Su Li,Elizabeth A. Mauldin,Susan Stellar,Marc De Meulder,Mark L. Kao,Daniel S. Ory,Cristin Davidson,Marie T. Vanier,Steven U. Walkley +18 more
TL;DR: These studies in a feline animal model have provided critical data on efficacy and safety of drug administration directly into the central nervous system that will be important for advancing HPβCD into clinical trials.
