Journal ArticleDOI
c-Jun Reprograms Schwann Cells of Injured Nerves to Generate a Repair Cell Essential for Regeneration
Peter Arthur-Farraj,Morwena Latouche,D Wilton,Susanne Quintes,Elodie Chabrol,Annbily Banerjee,Ashwin Woodhoo,Billy Jenkins,Mary Rahman,Mark Turmaine,Grzegorz Wicher,Richard Mitter,Linda Greensmith,Axel Behrens,Gennadij Raivich,Rhona Mirsky,Kristjan R. Jessen +16 more
TL;DR: It is concluded that a single glial transcription factor is essential for restoration of damaged nerves, acting to control the transdifferentiation of myelin and Remak Schwann cells to dedicated repair cells in damaged tissue.
Journal ArticleDOI
Notch controls embryonic Schwann cell differentiation, postnatal myelination and adult plasticity
Ashwin Woodhoo,Maria B Duran Alonso,Anna Droggiti,Mark Turmaine,Maurizio D'Antonio,David Parkinson,D Wilton,Raya Al-Shawi,Paul Simons,Jie Shen,François Guillemot,Freddy Radtke,Dies Meijer,M. Laura Feltri,Lawrence Wrabetz,Rhona Mirsky,Kristjan R. Jessen +16 more
TL;DR: Using transgenic mice and cell cultures, it is found that Notch has complex and extensive regulatory functions in Schwann cells, and this finding identifies a signaling pathway that induces myelin breakdown in vivo.
Journal ArticleDOI
Developmental alterations in Huntington's disease neural cells and pharmacological rescue in cells and mice
Ryan G. Lim,Lisa Salazar,D Wilton,Alvin R. King,Jennifer Stocksdale,Delaram Sharifabad,Alice L Lau,Beth Stevens,Jack C. Reidling,Sara T. Winokur,Malcolm Casale,Leslie M. Thompson,Mónica Pardo,A Gerardo García Díaz-Barriga,Marco Straccia,Phil Sanders,Jordi Alberch,Josep M. Canals,Julia A. Ross,Virginia B. Mattis,Colton M Tom,Loren Ornelas,Anais Sahabian,Lindsay Lenaeus,Berhan Mandefro,Dhruv Sareen,Clive N. Svendsen +50 more
TL;DR: The data suggest that mutant huntingtin impairs neurodevelopmental pathways that could disrupt synaptic homeostasis and increase vulnerability to the pathologic consequence of expanded polyglutamine repeat-associated phenotypes in neural cells and of cognitive impairment and synaptic pathology in HD model R6/2 mice.

Journal ArticleDOI
c-Jun activation in Schwann cells protects against loss of sensory axons in inherited neuropathy.
Janina Hantke,Lucy Carty,Laura J. Wagstaff,Mark Turmaine,D Wilton,Susanne Quintes,Martin Koltzenburg,Frank Baas,Rhona Mirsky,Kristjan R. Jessen +9 more
TL;DR: In this article, the authors showed that Schwann cells respond to injury by activating/enhancing trophic support for axons through a mechanism that depends on upregulation of the transcription factor c-Jun, resulting in the sparing of axons that would otherwise die.
