Journal ArticleDOI
Lewy bodies in grafted neurons in subjects with Parkinson's disease suggest host-to-graft disease propagation.
Jia-Yi Li,Elisabet Englund,Janice L. Holton,Denis Soulet,Peter Hagell,Andrew J. Lees,Tammaryn Lashley,Niall Quinn,Stig Rehncrona,Anders Björklund,Håkan Widner,Tamas Revesz,Olle Lindvall,Patrik Brundin +13 more
TL;DR: Two subjects with Parkinson's disease who had long-term survival of transplanted fetal mesencephalic dopaminergic neurons (11–16 years) developed α-synuclein–positive Lewy bodies in grafted neurons, providing the first evidence, to the authors' knowledge, that the disease can propagate from host to graft cells.
Journal ArticleDOI
Bone Marrow-Derived Microglia Play a Critical Role in Restricting Senile Plaque Formation in Alzheimer's Disease
TL;DR: This work shows a massive infiltration of highly ramified and elongated microglia within the core of amyloid plaques in transgenic mouse models of Alzheimer's disease (AD), and shows that blood-derivedmicroglia and not their resident counterparts have the ability to eliminate amyloids deposits by a cell-specific phagocytic mechanism.
Journal ArticleDOI
A novel pathogenic pathway of immune activation detectable before clinical onset in Huntington's disease
Maria Björkqvist,Edward J. Wild,Jenny Thiele,Aurelio Silvestroni,Ralph Andre,Nayana Lahiri,Elsa Raibon,Richard V. Lee,Caroline L. Benn,Denis Soulet,Anna K. Magnusson,Ben Woodman,Christian Landles,Mahmoud A. Pouladi,Michael R. Hayden,Azadeh Khalili-Shirazi,Mark W. Lowdell,Patrik Brundin,Gillian P. Bates,Blair R. Leavitt,Thomas Möller,Sarah J. Tabrizi +21 more
TL;DR: The peripheral immune system was examined and found widespread evidence of innate immune activation detectable in plasma throughout the course of HD, suggesting parallel central nervous system and peripheral pathogenic pathways of immune activation in HD.
Journal ArticleDOI
Platelets release mitochondria serving as substrate for bactericidal group IIA-secreted phospholipase A2 to promote inflammation
Luc H. Boudreau,Anne Claire Duchez,Nathalie Cloutier,Denis Soulet,Nicolas Martin,James G. Bollinger,Alexandre Paré,Matthieu Rousseau,Gajendra S. Naika,Tania Lévesque,Cynthia Laflamme,Geneviève Marcoux,Gérard Lambeau,Richard W. Farndale,Marc Pouliot,Hind Hamzeh-Cognasse,Fabrice Cognasse,Olivier Garraud,Peter A. Nigrovic,Helga Guderley,Steve Lacroix,Louis Thibault,John W. Semple,Michael H. Gelb,Eric Boilard +24 more
TL;DR: It is shown that activated platelets release respiratory-competent mitochondria, both within membrane-encapsulated microparticles and as free organelles, at the midpoint of a potent mechanism leading to inflammatory responses.
Journal ArticleDOI
Inhibition of Notch Signaling in Glioblastoma Targets Cancer Stem Cells via an Endothelial Cell Intermediate
Koos E. Hovinga,Fumiko Shimizu,Rong Wang,Georgia Panagiotakos,Maartje Van Der Heijden,Hamideh Moayedpardazi,Ana Sofia Correia,Ana Sofia Correia,Denis Soulet,Tamara Major,Jayanthi Menon,Viviane Tabar +11 more
TL;DR: A novel three‐dimensional organotypic “explant” system of surgical GBM specimens that preserves cytoarchitecture and tumor stroma along with tumor cells is used and suggests a critical role for tumor endothelial cells in GBM stem cell maintenance, mediated at least in part by Notch signaling.
