RNA velocity of single cells
Gioele La Manno,Gioele La Manno,Ruslan A. Soldatov,Amit Zeisel,Amit Zeisel,Emelie Braun,Emelie Braun,Hannah Hochgerner,Hannah Hochgerner,Viktor Petukhov,Viktor Petukhov,Katja Lidschreiber,Maria Eleni Kastriti,Peter Lönnerberg,Peter Lönnerberg,Alessandro Furlan,Jean Fan,Lars E. Borm,Lars E. Borm,Zehua Liu,David van Bruggen,Jimin Guo,Xiaoling He,Roger A. Barker,Erik Sundström,Gonçalo Castelo-Branco,Patrick Cramer,Patrick Cramer,Igor Adameyko,Sten Linnarsson,Sten Linnarsson,Peter V. Kharchenko +31 more
TL;DR: It is shown that RNA velocity—the time derivative of the gene expression state—can be directly estimated by distinguishing between unspliced and spliced mRNAs in common single-cell RNA sequencing protocols, and expected to greatly aid the analysis of developmental lineages and cellular dynamics, particularly in humans.
Journal ArticleDOI
Molecular Architecture of the Mouse Nervous System.
Amit Zeisel,Hannah Hochgerner,Peter Lönnerberg,Anna Johnsson,Fatima Memic,Job van der Zwan,Martin Häring,Emelie Braun,Lars E. Borm,Gioele La Manno,Simone Codeluppi,Alessandro Furlan,Kawai Lee,Nathan G. Skene,Kenneth D. Harris,Jens Hjerling-Leffler,Ernest Arenas,Patrik Ernfors,Ulrika Marklund,Sten Linnarsson +19 more
TL;DR: RNA sequencing of half a million single cells was used to create a detailed census of cell types in the mouse nervous system and mapped cell types spatially and derived a hierarchical, data-driven taxonomy.
Journal ArticleDOI
Molecular Diversity of Midbrain Development in Mouse, Human, and Stem Cells
Gioele La Manno,Gioele La Manno,Daniel Gyllborg,Simone Codeluppi,Simone Codeluppi,Kaneyasu Nishimura,Carmen Saltó,Amit Zeisel,Amit Zeisel,Lars E. Borm,Lars E. Borm,Simon R.W. Stott,Enrique M. Toledo,J. Carlos Villaescusa,J. Carlos Villaescusa,Peter Lönnerberg,Peter Lönnerberg,Jesper Ryge,Roger A. Barker,Ernest Arenas,Sten Linnarsson,Sten Linnarsson +21 more
TL;DR: Insight is provided into the molecular programs controlling human midbrain development and provides a foundation for the development of cell replacement therapies for Parkinson’s disease.

Molecular architecture of the mouse nervous system
Amit Zeisel,Hannah Hochgerner,Peter Lönnerberg,Anna Johnsson,Fatima Memic,Job van der Zwan,Martin Häring,Emelie Braun,Lars E. Borm,Gioele La Manno,Simone Codeluppi,Alessandro Furlan,Nathan G. Skene,Kenneth D. Harris,Jens Hjerling-Leffler,Ernest Arenas,Patrik Ernfors,Ulrika Marklund,Sten Linnarsson +18 more
TL;DR: RNA sequencing of half a million single cells is used to create a detailed census of cell types in the mouse nervous system and lays a solid foundation for understanding the molecular architecture of the mammalian nervous system, and enables genetic manipulation of specific cell types.
Journal ArticleDOI
Spatial organization of the somatosensory cortex revealed by osmFISH
Simone Codeluppi,Simone Codeluppi,Lars E. Borm,Lars E. Borm,Amit Zeisel,Amit Zeisel,Amit Zeisel,Gioele La Manno,Gioele La Manno,Josina Anna van Lunteren,Camilla I. Svensson,Sten Linnarsson,Sten Linnarsson +12 more
TL;DR: osmFISH applies automated cycles of single-molecule fluorescence in situ hybridization without barcoding to provide spatial gene expression in tissue sections at high sensitivity, accuracy and throughput.
