Journal ArticleDOI
Stearoyl-CoA desaturase 1 deficiency increases fatty acid oxidation by activating AMP-activated protein kinase in liver
Pawel Dobrzyn,Agnieszka Dobrzyn,Makoto Miyazaki,Paul Cohen,Esra Asilmaz,D. Grahame Hardie,Jeffrey M. Friedman,James M. Ntambi +7 more
TL;DR: The phosphorylation and activity of AMP-activated protein kinase (AMPK), a metabolic sensor that regulates lipid metabolism during increased energy expenditure is significantly increased and identified as a mediator of increased fatty acid oxidation in liver of SCD1-deficient mice.
Journal ArticleDOI
Site and mechanism of leptin action in a rodent form of congenital lipodystrophy
Esra Asilmaz,Paul Cohen,Makoto Miyazaki,Pawel Dobrzyn,Kohjiro Ueki,Gulnorakhon Fayzikhodjaeva,Alexander A. Soukas,C. Ronald Kahn,James M. Ntambi,Nicholas D. Socci,Jeffrey M. Friedman +10 more
TL;DR: It is shown that low-dose central leptin corrects the insulin resistance and fatty liver of lipodystrophic aP2-nSREBP-1c mice, while the same dose given peripherally does not, and data suggest that CNS-mediated repression of SCD-1 contributes to leptin's antisteatotic actions.
Journal ArticleDOI
Stearoyl-CoA desaturase 1 deficiency elevates insulin-signaling components and down-regulates protein-tyrosine phosphatase 1B in muscle.
Shaikh M. Rahman,Agnieszka Dobrzyn,Pawel Dobrzyn,Seong-Ho Lee,Makoto Miyazaki,James M. Ntambi +5 more
TL;DR: It is hypothesized that loss of SCD1 function induces increased insulin signaling at least in part by a reduction in the expression of protein-tyrosine phosphatase 1B, which could be a therapeutic target in the treatment of diabetes.
Book ChapterDOI
Mitochondria and Reactive Oxygen Species in Aging and Age-Related Diseases.
Carlotta Giorgi,Saverio Marchi,Ines C.M. Simoes,Ziyu Ren,Giampaolo Morciano,Mariasole Perrone,Paulina Patalas-Krawczyk,Sabine Borchard,Paulina Jędrak,Karolina Pierzynowska,Jędrzej Szymański,David Q.-H. Wang,Piero Portincasa,Grzegorz Węgrzyn,Hans Zischka,Pawel Dobrzyn,Massimo Bonora,Jerzy Duszyński,Alessandro Rimessi,Agnieszka Karkucinska-Wieckowska,Agnieszka Dobrzyn,Gyorgy Szabadkai,Barbara Zavan,Paulo J. Oliveira,Vilma A. Sardão,Paolo Pinton,Mariusz R. Wieckowski +26 more
TL;DR: An update regarding the key roles of ROS-mitochondria cross talk in different fundamental physiological or pathological situations accompanying aging is provided, highlighting that mitochondrial ROS may be a decisive target in clinical practice.
Journal ArticleDOI
Stearoyl-CoA desaturase-1 deficiency reduces ceramide synthesis by downregulating serine palmitoyltransferase and increasing β-oxidation in skeletal muscle
Agnieszka Dobrzyn,Pawel Dobrzyn,Seong-Ho Lee,Makoto Miyazaki,Paul Cohen,Esra Asilmaz,D. Grahame Hardie,Jeffrey M. Friedman,James M. Ntambi +8 more
TL;DR: Results indicate that SCD1 deficiency reduces ceramide synthesis by decreasing SPT expression and increasing the rate of beta-oxidation in oxidative muscles.
