TL;DR: Algorithms were developed to decipher this narrative and applied them to 21 breast cancers, finding that expansion of the dominant subclone to an appreciable mass may represent the final rate-limiting step in a breast cancer's development, triggering diagnosis.
Journal ArticleDOI
Analysis of the genetic phylogeny of multifocal prostate cancer identifies multiple independent clonal expansions in neoplastic and morphologically normal prostate tissue
Colin Cooper,Colin Cooper,Rosalind A. Eeles,Rosalind A. Eeles,David C. Wedge,Peter Van Loo,Peter Van Loo,Peter Van Loo,Gunes Gundem,Ludmil B. Alexandrov,Barbara Kremeyer,Adam Butler,Andy G. TL;DR: Genome-wide DNA sequencing was used to decrypt the phylogeny of multiple samples from distinct areas of cancer and morphologically normal tissue taken from the prostates of three men, demonstrating the existence of ongoing abnormal mutational processes, consistent with field effects, underlying carcinogenesis.
Journal ArticleDOI
Intra-tumour diversification in colorectal cancer at the single-cell level
Sophie F. Roerink,Nobuo Sasaki,Nobuo Sasaki,Henry Lee-Six,Matthew D. Young,Ludmil B. Alexandrov,Sam Behjati,Sam Behjati,Thomas J. Mitchell,Thomas J. Mitchell,Sebastian Grossmann,Howard Lightfoot,David A. Egan,Apollo Pronk,Niels Smakman,Joost van Gorp,Elizabeth Anderson,Stephen J. Gamble,Chris Alder,Marc van de Wetering,Peter J. Campbell,Michael R. Stratton,Hans Clevers +22 more
TL;DR: The results indicate that colorectal cancer cells experience substantial increases in somatic mutation rate compared to normal coloreCTal cells, and that genetic diversification of each cancer is accompanied by pervasive, stable and inherited differences in the biological states of individual cancer cells.
Journal ArticleDOI
Extensive transduction of nonrepetitive DNA mediated by L1 retrotransposition in cancer genomes

Janes,G. Steven Bova,Michael R. Stratton,Ultan McDermott,Peter J. Campbell,Peter J. Campbell +81 more
TL;DR: It is found that 3′ transduction activity in a patient’s tumor was always associated with hypomethylation of that element, and in some cases transduction events can scatter exons, genes, and regulatory elements widely across the genome.
Journal ArticleDOI

TL;DR: Exome sequencing in a series of 24 ACC cases identified multiple mutated genes that, combined, implicate chromatin deregulation in half of cases and identified the negative NOTCH signaling regulator, SPEN, as a new cancer gene in ACC with mutations in 5 cases.
