Journal ArticleDOI
Beta-secretase cleavage of Alzheimer's amyloid precursor protein by the transmembrane aspartic protease BACE.
Robert Vassar,Brian D. Bennett,Safura Babu-Khan,Steve Kahn,Elizabeth A. Mendiaz,Paul Denis,David B. Teplow,Sandra Ross,Patricia Amarante,Richard Loeloff,Yi Luo,Seth Fisher,Janis Fuller,Steven P. Edenson,Jackson Lile,Mark A. Jarosinski,Anja Leona Biere,Eileen Curran,Teresa L. Burgess,Jean Claude Louis,Frank H. Collins,James J. S. Treanor,Gary Rogers,Martin Citron +23 more
TL;DR: Overexpression of a transmembrane aspartic protease, termed BACE (for beta-site APP-cleaving enzyme) increased the amount of beta-secretase cleavage products, and these were cleaved exactly and only at known beta- secretase positions.
Journal ArticleDOI
GDNF–Induced Activation of the Ret Protein Tyrosine Kinase Is Mediated by GDNFR-α, a Novel Receptor for GDNF
Shuqian Jing,Duanzhi Wen,Yanbin Yu,Paige Holst,Yi Luo,Mei Fang,Rami Tamir,Laarni Antonio,Zheng Hu,Rod Cupples,Jean-Claude Louis,Sylvia Hu,Bruce W. Altrock,Gary M. Fox +13 more
TL;DR: In this paper, the expression cloning and characterization of GDNFR-α, a novel glycosylphosphatidylinositol-linked cell surface receptor for glial cell line-derived neurotrophic factor (GDNF), was reported.
Journal ArticleDOI
Mice deficient in BACE1, the Alzheimer's beta-secretase, have normal phenotype and abolished beta-amyloid generation.
Yi Luo,Brad Bolon,Steve Kahn,Brian D. Bennett,Safura Babu-Khan,Paul Denis,Wei Fan,Hue Kha,Jianhua Zhang,Yunhua Gong,Laura Martin,Jean Claude Louis,Qiao Yan,William G. Richards,Martin Citron,Robert Vassar +15 more
TL;DR: These results provide validation of BACE1 as the major β-secretase in vivo and suggest that therapeutic inhibition of Bace1 for the treatment of Alzheimer's disease may be free of mechanism-based toxicity.
Journal ArticleDOI
Neu differentiation factor: a transmembrane glycoprotein containing an EGF domain and an immunoglobulin homology unit.
Duanzhi Wen,Elior Peles,Rod Cupples,Sidney Vaughn Suggs,Sarah S. Bacus,Yi Luo,Geraldine Trail,Sylvia Hu,Scott M. Silbiger,Rachel Levy,Raymond A. Koski,Hsieng Sen Lu,Yosef Yarden +12 more
TL;DR: The deduced structure of the precursor of NDF indicated that it is a transmembrane protein whose extracellular portion contains an EGF-like domain that probably functions as a receptor recognition site and is released into the medium of transfected COS-7 cells in a biologically active form.
Journal ArticleDOI
Structural and functional aspects of the multiplicity of Neu differentiation factors.
Duanzhi Wen,Sidney Vaughn Suggs,Devarajan Karunagaran,N Liu,Rod Cupples,Yi Luo,A M Janssen,N Ben-Baruch,D B Trollinger,V L Jacobsen +9 more
TL;DR: Results suggest that different NDF isoforms are generated by alternative splicing and perform distinct tissue-specific functions.
