Abstract: The Wnt signaling pathway is conserved in various species from worms to mammals, and plays important roles in cellular proliferation, differentiation, and migration. Wnt stabilizes cytoplasmic beta-catenin and then the accumulated beta-catenin is translocated into the nucleus, where it activates the transcriptional factor T-cell factor (Tcf)/lymphoid enhancer factor (Lef), and thereby stimulates the expression of genes including c-myc, c-jun, fra-1, and cyclin D1. Tight regulation of this response involves post-translational modifications of the components of the Wnt signaling pathway. Phosphorylation, ubiquitination, and sumoylation have been shown to affect the half-life of beta-catenin and the transcriptional activity of Tcf/Lef. The precise spatio-temporal patterns of these multiple modifications determine the driving force of various cellular responses.

Abstract: Green space is particularly indispensable for proper functioning of the ecosystem in an urban environment. This study was an attempt to dynamically map and monitor green spaces in Greater Dhaka of Bangladesh. Both primary and secondary data were acquired to document the spatial–temporal dynamics of green spaces in the study area. Using a supervised classification algorithm, multi-temporal land use/cover data were extracted from a set of satellite images. A number of spatial metrics were employed to understand the landscape condition in a multi-temporal manner. In addition, 50 key informants along with focus group discussion and observation techniques were used to document existing management aspects of green spaces and their conservation policies. The analysis revealed that green spaces in Greater Dhaka are rapidly disappearing over the course of time even though they provide a number of natural, economic and social benefits. The disappearance of green spaces was primarily attributed to a rapid increase in the urban population, mainly driven by rural–urban migration. As a result, the landscape became highly fragmented and less connected. A substantial reduction of green patches is also leading to deterioration of the ecological condition of the landscape. The drastic reduction of green spaces in Greater Dhaka has been attributed to a lack of policy, low political motivation, and poor management. In order to ensure sustainability of green spaces and proper functioning of the city’s ecosystem, there is an urgent need for strategic green space planning.

Abstract: Transcription activator-like effector (TALE) nuclease (TALEN) is a site-specific nuclease, which can be freely designed and easily constructed Numerous methods of constructing TALENs harboring different TALE scaffolds and repeat variants have recently been reported However, the functionalities of structurally different TALENs have not yet been compared Here, we report on the functional differences among several types of TALENs targeting the same loci Using HEK293T cell-based single-strand annealing and Cel-I nuclease assays, we found that TALENs with periodically-patterned repeat variants harboring non-repeat-variable di-residue (non-RVD) variations (Platinum TALENs) showed higher activities than TALENs without non-RVD variations Furthermore, the efficiencies of gene disruption mediated by Platinum TALENs in frogs and rats were significantly higher than in previous reports This study therefore demonstrated an efficient system for the construction of these highly active Platinum TALENs (Platinum Gate system), which could establish a new standard in TALEN engineering
