Abstract: Objective and Design: We investigated the effect of a new class of COX-2 inhibitor, rutaecarpine, on the production of PGD2 in bone marrow derived mast cells (BMMC) and PGE2 in COX-2 transfected HEK293 cells Inflammation was induced by γ-carrageenan in male Splague-Dawley (SD) rats¶Material: Rutaecarpine (8,13-Dihydroindolo[2',3':3,4]pyridol[2,1-b]quinazolin-5(7H)-one) was isolated from the fruits of Evodia rutaecarpa BMMC were cultured with WEHI-3 conditioned medium c-Kit ligand and IL-10 were obtained by their expression in baculovirus¶Methods: The generation of PGD2 and PGE2 were determined by their assay kit COX-1 and COX-2 protein and mRNA expression was determined by BMMC in the presence of KL, LPS and IL-10¶Treatment: Rutaecarpine and indomethacin dissolved in 01% carboxymethyl cellulose was administered intraperitoneally and, 1h later, γ-carrageenan solution was injected to right hind paw of rats Paw volumes were measured using plethysmometer 5h after γ-carrageenan injection¶Results: Rutaecarpine inhibited COX-2 and COX-1 dependent phases of PGD2 generation in BMMC in a concentration-dependent manner with an IC50 of 028 μM and 87 μM, respectively It inhibited COX-2-dependent conversion of exogenous arachidonic acid to PGE2 in a dose-dependent manner by the COX-2-transfected HEK293 cells However, rutaecarpine inhibited neither PLA2 and COX-1 activity nor COX-2 protein and mRNA expression up to the concentration of 30 μM in BMMC, indicating that rutaecarpine directly inhibited COX-2 activity Furthermore, rutaecarpine showed in vivo anti-inflammatory activity on rat γ-carrageenan induced paw edema by intraperitoneal administration¶Conclusion: Anti-inflammatory activity of Evodia rutaecarpa could be attributed at least in part by inhibition of COS-2

Abstract: A measurement is presented of W-boson production in PbPb collisions carried out at a nucleon-nucleon (NN) centre-of-mass energy sqrt(s[NN]) of 2.76 TeV at the LHC using the CMS detector. In data corresponding to an integrated luminosity of 7.3 inverse microbarns, the number of W to mu mu-neutrino decays is extracted in the region of muon pseudorapidity abs(eta[mu]) 25 GeV. Yields of muons found per unit of pseudorapidity correspond to (159 +/- 10 (stat.) +/- 12 (syst.)) 10E-8 W(plus) and (154 +/- 10 (stat.) +/- 12 (syst.)) 10E-8 W(minus) bosons per minimum-bias PbPb collision. The dependence of W production on the centrality of PbPb collisions is consistent with a scaling of the yield by the number of incoherent NN collisions. The yield of W bosons is also studied in a sample of pp interactions at sqrt(s)= 2.76 TeV corresponding to an integrated luminosity of 231 inverse nanobarns. The individual W(plus) and W(minus) yields in PbPb and pp collisions are found to agree, once the neutron and proton content in Pb nuclei is taken into account. Likewise, the difference observed in the dependence of the positive and negative muon production on pseudorapidity is consistent with next-to-leading order perturbative QCD calculations.

Abstract: Although approximately 25 common genetic susceptibility loci have been identified to be independently associated with breast cancer risk through genome-wide association studies (GWAS), the genetic risk variants reported to date only explain a small fraction of the heritability of breast cancer. Furthermore, GWAS-identified loci were primarily identified in women of European descent. To evaluate previously identified loci in Korean women and to identify additional novel breast cancer susceptibility variants, we conducted a three-stage GWAS that included 6,322 cases and 5,897 controls. In the validation study using Stage I of the 2,273 cases and 2,052 controls, seven GWAS-identified loci [5q11.2/MAP3K1 (rs889312 and rs16886165), 5p15.2/ROPN1L (rs1092913), 5q12/MRPS30 (rs7716600), 6q25.1/ESR1 (rs2046210 and rs3734802), 8q24.21 (rs1562430), 10q26.13/FGFR2 (rs10736303), and 16q12.1/TOX3 (rs4784227 and rs3803662)] were significantly associated with breast cancer risk in Korean women (Ptrend < 0.05). To identify additional genetic risk variants, we selected the most promising 17 SNPs in Stage I and replicated these SNPs in 2,052 cases and 2,169 controls (Stage II). Four SNPs were further evaluated in 1,997 cases and 1,676 controls (Stage III). SNP rs13393577 at chromosome 2q34, located in the Epidermal Growth Factor Receptor 4 (ERBB4) gene, showed a consistent association with breast cancer risk with combined odds ratios (95% CI) of 1.53 (1.37-1.70) (combined P for trend = 8.8 × 10-14). This study shows that seven breast cancer susceptibility loci, which were previously identified in European and/or Chinese populations, could be directly replicated in Korean women. Furthermore, this study provides strong evidence implicating rs13393577 at 2q34 as a new risk variant for breast cancer.
