Abstract: Introduction: Platelet-rich plasma contains autologous thrombocyte growth factors and might be promising for acceleration of dentoalveolar bone regeneration. In this study, it was analysed for platelet counts and growth factor concentrations. Material and method: Platelet-rich plasma was isolated by discontinuous cell separation from 158 healthy men and 55 women aged 17–62 years. One hundred and fifteen specimens (stratified for age and gender of the donor) were analysed for growth factor concentrations and platelet count. Results: The platelet count in platelet-rich plasma (1,407,640±320,100/μl) was 5 times higher than in donor blood (266,040±60,530/μl). Platelet-derived growth factor AB (117±63 ng/ml), transforming growth factor (TGF) β -1 (169±84 ng/ml), and insulin-like growth factor (IGF) I (84±23 ng/ml) were found in large amounts, while platelet-derived growth factor (PDGF) BB (10±8 ng/ml) and transforming growth factor β -2 (0.4±0.3 ng/ml) were found in small amounts only. The growth factor content was not well correlated with the platelet count in whole blood nor with the platelet-rich plasma ( r p =0.35). No influence of gender or age on platelet count or growth factor concentrations was discovered (except IGF-I). Conclusions: While there was substantial variation in the growth factor content of platelet-rich plasma, the factors influencing this are still worthy of further investigation. Furthermore, a technique whereby the growth factor content could be rapidly assessed in platelet-rich plasma may be of therapeutic benefit. Abstract: We introduce and propose zircon M257 as a future reference material for the determination of zircon U-Pb ages by means of secondary ion mass spectrometry. This light brownish, flawless, cut gemstone specimen from Sri Lanka weighed 5.14 g (25.7 carats). Zircon M257 has TIMS-determined, mean isotopic ratios (2s uncertainties) of 0.09100 ± 0.00003 for 206pb/238U and 0.7392 ± 0.0003 for 207pb/235U. Its 206pb/238U age is 561.3 ± 0.3 Ma (unweighted mean, uncertainty quoted at the 95% confidence level); the U-Pb system is concordant within uncertainty of decay constants. Zircon M257 contains ∼ 840 μg g−1 U (Th/U ∼ 0.27). The material exhibits remarkably low heterogeneity, with a virtual absence of any internal textures even in cathodoluminescence images. The uniform, moderate degree of radiation damage (estimated from the expansion of unit-cell parameters, broadening of Raman spectral parameters and density) corresponds well, within the “Sri Lankan trends”, with actinide concentrations, U-Pb age, and the calculated alpha fluence of 1.66 × 1018 g−1. This, and a (U+Th)/He age of 419 ± 9 Ma (2s), enables us to exclude any unusual thermal history or heat treatment, which could potentially have affected the retention of radiogenic Pb. The oxygen isotope ratio of this zircon is 13.9%o VSMOW suggesting a metamorphic genesis in a marble or calc-silicate skarn. Nous presentons et proposons a la communaute le zircon M257 qui pourrait devenir un materiau de reference pour la determination des âges U-Pb sur zircon par spectrometrie de masse a ions secondaires (SIMS). Ce specimen scie d'une gemme brun clair, sans defauts, provenant du Sri Lanka, pese 5.14 g (25.7 carats). Les moyennes des rapports isotopiques (et les incertitudes associees 2s) mesurees par TIMS sur ce zircon M257 sont 0.09100 ± 0.00003 en 206pb/238U et 0.7392 ± 0.0003 en 207pb/235U. Son âge 206pb/238U est de 561.3 ± 0.3 Ma (moyenne non ponderee, incertitude: 95% intervalle de confiance); le systeme U-Pb y est concordant, aux erreurs sur les constantes de desintegration pres. Le zircon M257 contient ∼ 840 μg g−1 U (Th/U ∼ 0.27). Ce materiel montre de tres faibles heterogeneites, avec une quasi absence de textures internes, meme a I'examen par cathodoluminescence. Le degre de dommage lie aux radiations (estime par I'expansion des parametres de la cellule unitaire, I'elargissement des parametres des spectres Raman et la densite) est modere et uniforme. II se correle bien, (sur I'alignement “Sri Lanka”) avec les concentrations en actinides, I'âge U-Pb et la fluence calculee de 1.66 × 1018 g−1 Ceci, couplea son âge (U+Th)/He de 419 ± 9 Ma (2s), nous permet d'exclure I'existence d'evenement thermique passe, ou de traitement par la chaleur, qui aurait potentiellement pu affecter le processus de retention de Pb radiogenique. Le rapport isotopique de I'oxygene de ce zircon est 13.9%o VSMOW, suggerant une genese par metamorphisme, au sein d'un marbre ou d'un skarn calco-silicate.

Abstract: Summary Background Graft-versus-host disease (GVHD) is a major cause of morbidity and mortality after allogeneic haematopoietic cell transplantation from unrelated donors. Anti-T-cell globulins (ATGs) might lower the incidence of GVHD. We did a prospective, randomised, multicentre, open-label, phase 3 trial to compare standard GVHD prophylaxis with ciclosporin and methotrexate with or without anti-Jurkat ATG-Fresenius (ATG-F). Methods Between May 26, 2003, and Feb 8, 2007, 202 patients with haematological malignancies were centrally randomly assigned using computer-generated centre-stratified block randomisation between treatment groups receiving ciclosporin and methotrexate with or without additional ATG-F. One patient in the ATG-F group did not undergo transplantation, thus 201 patients who underwent transplantation with peripheral blood (n=164; 82%) or bone marrow (n=37; 18%) grafts from unrelated donors after myeloablative conditioning were included in the full analysis set, and were analysed according to their randomly assigned treatment (ATG-F n=103, control n=98). The primary endpoint was severe acute GVHD (aGVHD) grade III–IV or death within 100 days of transplantation. The trial is registered with the numbers DRKS00000002 and NCT00655343. Findings The number of patients in the ATG-F group who had severe aGVHD grade III–IV or who died within 100 days of transplantation was 12 and 10 (21·4%, 95% CI 13·4–29·3), respectively, compared with 24 and nine (33·7%, 24·3–43·0) patients, respectively, in the control group (adjusted odds ratio 0·59, 95% CI 0·30–1·17; p=0·13). The cumulative incidence of aGVHD grade III–IV was 11·7% (95% CI 6·8–19·8) in the ATG-F group versus 24·5% (17·3–34·7) in the control group (adjusted hazard ratio [HR] 0·50, 95% CI 0·25–1·01; p=0·054), and cumulative incidence of aGVHD grade II–IV was 33·0% (n=34; 95% CI 25·1–43·5) in the ATG-F group versus 51·0% (n=50; 95% CI 42·0–61·9) in the control group (adjusted HR 0·56, 0·36–0·87; p=0·011). The 2-year cumulative incidence of extensive chronic GVHD was 12·2% (n=11; 95% CI 7·0–21·3) versus 42·6% (n=34; 95% CI 33·0–55·0; adjusted HR 0·22, 0·11–0·43; p Interpretation The addition of ATG-F to GVHD prophylaxis with ciclosporin and methotrexate resulted in decreased incidence of acute and chronic GVHD without an increase in relapse or non-relapse mortality, and without compromising overall survival.
