Discovery of SHR2415, a Novel Pyrrole-Fused Urea Scaffold ERK1/2 Inhibitor.

TL;DR: A novel scaffold featuring a pyrrole-fused urea template was designed and synthesized and the lead compound, SHR2415, was shown to be a highly potent ERK1/2 inhibitor that exhibited high cell potency based on the Colo205 assay.

Improving the predictive power of xenograft and syngeneic anti-tumour studies using mice humanised for pathways of drug metabolism

TL;DR: A complex transgenic mouse model – 8HUM - is developed which faithfully replicates human Phase I drug metabolism (and its regulation), and which will generate more human-relevant data from fewer animals in a pre-clinical setting and reduce attrition in the clinic.
Journal Article

New drugs in development for melanoma.

TL;DR: Despite the enormous progress over the past 5 or 6 years in developing new agents that substantially prolong life in metastatic disease, the majority of patients with metastatic melanoma still die of their disease, and there is a high rate of resistance to melanoma treatments.

Cutaneous Melanoma

- 01 Jan 2022 -
TL;DR: In this paper , the authors addressed the topic of melanoma in children and found that the benefits of new therapies for adult melanoma have not been reflected in younger patients, and more efforts are needed to facilitate the transfer of potentially effective new agents from adults to children and to improve research programs dedicated to young melanoma patients.

A multiple testing procedure for clinical trials.

TL;DR: The overall size of the procedure is shown to be controlled with virtually the same accuracy as the single sample chi-square test based on N(m1 + m2) observations and the power is found to bevirtually the same.
