This protocol is for the in-gel digestion of both silver and Coomassie-stained protein spots or bands and can be followed by MALDI-MS or LC-MS/MS analysis to identify proteins at sensitivities better than a few femtomoles of protein starting material.Abstract:
In-gel digestion of proteins isolated by gel electrophoresis is a cornerstone of mass spectrometry (MS)-driven proteomics. The 10-year-old recipe by Shevchenko et al. has been optimized to increase the speed and sensitivity of analysis. The protocol is for the in-gel digestion of both silver and Coomassie-stained protein spots or bands and can be followed by MALDI-MS or LC-MS/MS analysis to identify proteins at sensitivities better than a few femtomoles of protein starting material.read more

A method for rapid production of heteromultimeric protein complexes in plants: assembly of protective bluetongue virus-like particles.
TL;DR: The results show that transient expression can be used to produce immunologically relevant complex heteromultimeric structures in plants in a matter of days and have implications beyond the realm of veterinary vaccines.

ER stress potentiates insulin resistance through PERK-mediated FOXO phosphorylation
Wei Zhang,Ville Hietakangas,Sheena Wee,Siew Choo Lim,Jayantha Gunaratne,Stephen M. Cohen,Stephen M. Cohen +6 more
TL;DR: Evidence is presented that the ER stress pathway acts via a conserved signaling mechanism involving the protein kinase PERK to modulate cellular insulin responsiveness, and it is suggested that PERK may be a promising pharmacological target for ameliorating insulin resistance.

Detergent selection for enhanced extraction of membrane proteins.
TL;DR: Different detergents were more effective than others in the extraction of specific classes of integral membrane proteins, offering guidelines for initial detergent selection and supports the value of detergent screening and optimization to increase the yields of targeted membrane proteins.

Systemic Cold Stress Adaptation of Chlamydomonas reinhardtii
TL;DR: Cold stress adaptation of C. reinhardtii is studied to integrate proteome, metabolome, physiological and cell-morphological changes during a time-course from 0 to 120 h and revealed correlations between proteins and metabolites otherwise not detectable.

Mass Spectrometric Sequencing of Proteins from Silver-Stained Polyacrylamide Gels
TL;DR: Silver staining allows a substantial shortening of sample preparation time and may, therefore, be preferable over Coomassie staining, and this work removes a major obstacle to the low-level sequence analysis of proteins separated on polyacrylamide gels.

TL;DR: Two recently developed methodologies offer the opportunity to obtain quantitative proteomic information by comparing the signals from the same peptide under different conditions, and stable isotope labels facilitates direct quantification from the mass spectra.
