TLDR
Modified long composite primers were developed based on both mRNA differential display and RNA arbitrarily primed PCR fingerprinting methods to increase the reproducibility and sensitivity of the mRNA differentialdisplay while still keeping the characteristics of the original method.
Abstract
To increase the reproducibility and to reduce the false positives in the initial mRNA differential display, modified long composite primers were developed based on both mRNA differential display and RNA arbitrarily primed PCR fingerprinting methods. Ten-base nucleotides were added at the 5' ends of the primers used in the initial mRNA differential display. These included a restriction site to aid cloning. PCR began with one low-stringency cycle (40 degrees C for annealing) followed by 35 high-stringency cycles (60 degrees C for annealing). The modified method significantly improved the reproducibility and sensitivity of the mRNA differential display while still keeping the characteristics of the original method.

Tamoxifen Down-Regulates CD36 Messenger RNA Levels in Normal and Neoplastic Human Breast Tissues

TL;DR: Results indicate that the down-regulation of CD36 induced by TAM might represent alternative or additional mechanisms of action of this drug affecting the functions of thrombospondin-1, which is involved in hematogenous tumor spread, invasion and angiogenesis, and oxidized low-density lipoprotein, playing a role in inhibition of arteriosclerosis.

In search of differentially expressed genes and proteins

TL;DR: It is shown how data from RNA/DNA mapping, sequence information from genome projects and protein pattern profiling can be linked with each other and annotated and permit the study of differential gene and protein expressions in cells or tissues.

Universal RT-coupled PCR method for the specific amplification of mRNA

TL;DR: In this paper, a novel Universal RT-coupled PCR strategy for the specific detection and accurate quantification of mRNA was proposed, and a specific primer mix containing the universal RT-primers, a transcript specific forward primer and a reverse PCR primer identical to a unique tag sequence.

Identification of a novel gene, DAM1, amplified at chromosome 1p13.3-21 region in human breast cancer cell lines.

TL;DR: The findings suggest that the DAM1 gene is a novel gene up-regulated by amplification in human breast cancer cell lines, and is associated with gene amplification in MCF-7 and BT-20 breast cancer cells.

Identification and cloning of differentially expressed genes by long-distance differential display.

TL;DR: Coupled with high-throughput cDNA sequencing and multiplex hybridization of cDNA microarrays for confirmation of differential expression, LDD-PCR could prove to be useful for simultaneous scanning of transcripts from multiple cDNA samples and faster selection of differentially expressed transcripts of interest.
