TLDR
Evidence is presented indicating that only one tetrameric repressor molecule binds strongly to a segment of four (or fewer) tandem operators, but that two repressor molecules can be accommodated on segments containing at least six tandem operators.

The tac promoter: a functional hybrid derived from the trp and lac promoters

TL;DR: Two hybrid promoters that are functional in Escherichia coli have been constructed and are useful for the controlled expression of foreign genes at high levels in E. coli.

Diffusion-Driven Mechanisms of Protein Translocation on Nucleic Acids. 1. Models and Theory?

TL;DR: Four types of processes that may be involved in protein translocation events between DNA sites are defined, and the consequences of each for the overall rate of target location are worked out as a function of both the nonspecific binding affinity between protein and DNA and the length of the DNA molecule containing the target sequence.
Journal ArticleDOI

Ty elements transpose through an RNA intermediate

TL;DR: It is demonstrated that during transposition, Ty sequence information flows from DNA to RNA to DNA, and the pattern of sequence inheritance in progeny Ty insertions derived from the marked Ty follows the predictions of the model of retroviral reverse transcription.

Formation of de novo centromeres and construction of first-generation human artificial microchromosomes.

TL;DR: This first-generation system for the construction of human artificial chromosomes should be suitable for dissecting the sequence requirements of human centromeres, as well as developing constructs useful for therapeutic applications.
Journal ArticleDOI

Structure, sequence and expression of the hepatitis delta ( δ ) viral genome

TL;DR: Biochemical and electron microscopic data indicate that the human hepatitis δ viral agent contains a covalently closed circular and single-stranded RNA genome that has certain similarities with viroid-like agents from plants.

SUPERCOILED CIRCULAR DNA-PROTEIN COMPLEX IN Escherichia coli: PURIFICATION AND INDUCED CONVERSION TO AN OPEN CIRCULAR DNA FORM

TL;DR: Electron microscopic analyses indicate that the decrease in sedimentation rate of the ColE(1)-protein complex after treatment with these various agents is largely owing to an induced transition of ColE (1) DNA from the supercoiled to the open circular state.

Construction and characterization of new cloning vehicles. I. Ampicillin-resistant derivatives of the plasmid pMB9.

TL;DR: In vitro recombination via restriction endonucleases and the in vivo genetic translocation of the Ap resistance (Apr) gene resulted in the construction of a new cloning vehicle, the plasmid pBR313, which has a molecular weight of 5.8 Mdalton and has been characterized using thirteen restriction enzymes, six of which cleave the plasid at unique restriction sites.

Plasmid ColE1 as a Molecular Vehicle for Cloning and Amplification of DNA

TL;DR: DNA fragments obtained from EcoRI endonuclease digestion of bacteriophage varphi80pt190 (trp(+)) and the plasmid ColE1 were covalently joined with polynucleotide ligase to produce plasmids containing the trp operon (ColE1-trp) and the kanamycin resistance gene.
