Electron tomography : methods for three-dimensional visualization of structures in the cell
TL;DR: The principles of electron microscopy have been discussed in this article, including the use of the Electron Microscope as a structure projector, and the role of the Markerless Alignment in Electron Tomography.
Journal ArticleDOI
Electron cryo-microscopy shows how strong binding of myosin to actin releases nucleotide
TL;DR: By fitting atomic models of actin and the myosin cross-bridge into high-resolution electron cryo-microscopy three-dimensional reconstructions, the molecular basis of this linkage is described and the closing of the actin-binding cleft when actin binds is structurally coupled to the opening of the nucleotide-binding pocket.
Journal ArticleDOI
Optimizing phase contrast in transmission electron microscopy with an electrostatic (Boersch) phase plate.
E. Majorovits,Bastian Barton,K. Schultheiß,Fabian Perez-Willard,Dagmar Gerthsen,Rasmus R. Schröder +5 more
TL;DR: The first fabrication and application of a miniaturized electrostatic einzel lens driven as TEM quarter-wave phase plate, which proves to be mechanically stable and does not impair image quality, in particular it does not reduce the high-resolution signal.
Book ChapterDOI
Image restoration in cryo-electron microscopy
TL;DR: It is shown that the Wiener filter-based methodology can be used to obtain a solution to the problem of amplitude correction (or "sharpening") of the EM map that makes it visually comparable to maps determined by X-ray crystallography, and thus amenable to comparative interpretation.
Journal ArticleDOI
Electron energy filtering significantly improves amplitude contrast of frozen-hydrated protein at 300 kV
TL;DR: Energy filtering effectively doubled the signal-to-noise ratio in the images of frozen-hydrated filaments, and substantially improved intensity data statistics of layer lines up to at least approximately 25A resolution in their Fourier transforms.

The use of fast Fourier transform for the estimation of power spectra: A method based on time averaging over short, modified periodograms
TL;DR: In this article, the use of the fast Fourier transform in power spectrum analysis is described, and the method involves sectioning the record and averaging modified periodograms of the sections.

Transmission Electron Microscopy
TL;DR: The client would like to get a larger, approximately 3 cm in diameter, well fixed tissue sample, together with a detailed report of the clinical presentation, gross, and microscopic lesions, along with the submission of samples prepared in a similar manner by the client for processing.
Journal ArticleDOI
Model for the structure of bacteriorhodopsin based on high-resolution electron cryo-microscopy.
TL;DR: A complete atomic model for bacteriorhodopsin between amino acid residues 8 and 225 has been built and suggests that pK changes in the Schiff base must act as the means by which light energy is converted into proton pumping pressure in the channel.

TL;DR: Water is the most abundant component of biological material, but it is systematically excluded from conventional electron microscopy, because water evaporates rapidly under the vacuum conditions of an electron microscope.
