An ACT1 Mutation Selectively Abolishes Interleukin-17 Responses in Humans with Chronic Mucocutaneous Candidiasis

TL;DR: It is reported that a biallelic missense mutation (T536I) in the adaptor molecule ACT1 in two siblings with CMC abolished the homotypic interaction of ACT1 with IL-17 receptors, with no effect on homodimerization, and other ACT1-dependent IL- 17 cytokines seem to be largely redundant in host defense.

Act1 mediates IL-17–induced EAE pathogenesis selectively in NG2 + glial cells

TL;DR: NG2+ glia is identified as the major CNS cellular target of IL-17 in EAE, and the sensitivity of oligodendrocyte lineage cells to IL- 17–mediated toxicity further suggests a direct link between inflammation and neurodegeneration in multiple sclerosis.
