TL;DR: It is demonstrated that patients with hypomorphic RAG mutations, especially those with delayed-onset combined immune deficiency and granulomatous/autoimmune manifestations (CID-G/AI), produce a broad spectrum of autoantibodies and indicates that environmental triggers may modulate the phenotypic expression of autoimmune manifestations.

TL;DR: By conditional gene deletion, mice with selective deficiency of WASp in the B-cell lineage are generated and it is shown that this is sufficient to cause a severe reduction of marginal zone B cells and inability to respond to type II T-independent Ags, thereby recapitulating phenotypic features of complete WASp deficiency.

TL;DR: In this article, a population genetic analysis was performed to estimate the incidence of disease caused by RAG1/2 homozygous or compound heterozygous mutations and found that the contribution of immune dysregulatory disease due to RAG 1/2 mutations present in the general population may be much higher than previously estimated.
