Journal ArticleDOI
Universal cardiac induction of human pluripotent stem cells in two and three-dimensional formats: implications for in vitro maturation.
Miao Zhang,Jan S. Schulte,Alexander Heinick,Ilaria Piccini,Jyoti Rao,Roberto Quaranta,Dagmar Zeuschner,Daniela Malan,Kee-Pyo Kim,Albrecht Röpke,Philipp Sasse,Marcos J. Araúzo-Bravo,Guiscard Seebohm,Hans R. Schöler,Larissa Fabritz,Paulus Kirchhof,Frank U. Müller,Boris Greber +17 more
TL;DR: In this paper, the authors used systematic combinatorial optimization to narrow down the key requirements for efficient cardiac induction of human pluripotent stem cells (hPSCs) in 2D and 3D culture formats.
Journal ArticleDOI
Overexpression of cAMP-response element modulator causes abnormal growth and development of the atrial myocardium resulting in a substrate for sustained atrial fibrillation in mice
Paulus Kirchhof,Eloi Marijon,Larissa Fabritz,Na Li,Wei Wang,Tiannan Wang,Kirsten Schulte,Juliane Hanstein,Jan S. Schulte,Mathis Vogel,Nathalie Mougenot,Sandra Laakmann,Lisa Fortmueller,Jens Eckstein,Sander Verheule,Sven Kaese,Ariane Staab,Stephanie Grote-Wessels,Ulrich Schotten,Ghassan Moubarak,Xander H.T. Wehrens,Wilhelm Schmitz,Stéphane N. Hatem,Frank U. Müller +23 more
TL;DR: It is concluded that transcription factor CREM is an important regulator of atrial growth implicated in the development of an arrhythmogenic substrate in TG mice.
Journal ArticleDOI
Sphingosine‐1‐Phosphate Receptor 1 Regulates Cardiac Function by Modulating Ca2+ Sensitivity and Na+/H+ Exchange and Mediates Protection by Ischemic Preconditioning
Petra Keul,Marcel M. G. J. van Borren,Alexander Ghanem,Frank Müller,Antonius Baartscheer,Arie O. Verkerk,Frank Stümpel,Jan S. Schulte,Nazha Hamdani,Wolfgang A. Linke,Pieter B. van Loenen,Marek Matus,Wilhelm Schmitz,Jörg Stypmann,Klaus Tiemann,Jan-Hindrik Ravesloot,Astrid E. Alewijnse,Sven Hermann,Léon J. A. Spijkers,Karl-Heinz Hiller,Deron R. Herr,Gerd Heusch,Michael Schäfers,Stephan L. M. Peters,Jerold Chun,Bodo Levkau +25 more
TL;DR: Tonic S1P1 signaling by endogenous sphingosine‐1‐phosphate contributes to intracellular Ca2 + homeostasis by maintaining basal NHE‐1 activity and controls simultaneously myofibril Ca2+ sensitivity through its inhibitory effect on adenylate cyclase.
Journal ArticleDOI
Cardiac function is regulated by B56α-mediated targeting of protein phosphatase 2A (PP2A) to contractile relevant substrates.
Uwe Kirchhefer,Christiane Brekle,John Eskandar,Gunnar Isensee,Dana Kucerova,Frank Müller,Florence Pinet,Jan S. Schulte,Matthias D. Seidl,Peter Boknik +9 more
TL;DR: Cardiomyocyte-directed overexpression of B56α, the main cardiac PP2A regulatory subunit, results in the dephosphorylation of myofilament proteins, increased Ca2+ sensitivity, and higher contractility, which suggests an important role for B56 α in regulating PP1A activity and thereby the contractile function.
Journal ArticleDOI
Stretch-activated two-pore-domain (K2P) potassium channels in the heart: Focus on atrial fibrillation and heart failure.
Constanze Schmidt,Felix Wiedmann,Stefan Kallenberger,Antonius Ratte,Jan S. Schulte,Beatrix Scholz,Frank U. Müller,Niels Voigt,Maria-Patapia Zafeiriou,Joachim R. Ehrlich,Ursula Tochtermann,Gábor Veres,Arjang Ruhparwar,Matthias Karck,Hugo A. Katus,Dierk Thomas +15 more
TL;DR: Assessment of expression and remodeling of mechanosensitive K2P channels in atrial fibrillation (AF) and heart failure (HF) patients in comparison to murine models suggested a mechanistic contribution to cardiac arrhythmogenesis.
