Generation of Gene-Modified Cynomolgus Monkey via Cas9/RNA-Mediated Gene Targeting in One-Cell Embryos

TL;DR: By coinjection of Cas9 mRNA and sgRNAs into one-cell-stage embryos, this system successfully achieves precise gene targeting in cynomolgus monkeys and enables simultaneous disruption of two target genes in one step, and no off-target mutagenesis was detected by comprehensive analysis.

Anti-tumour immunity controlled through mRNA m 6 A methylation and YTHDF1 in dendritic cells

TL;DR: It is shown that durable neoantigen-specific immunity is regulated by mRNA N6-methyadenosine (m6A) methylation through the m6A-binding protein YTHDF15, which suppresses the clearance of tumours by enhancing the translation of lysosomal proteases in dendritic cells and thereby suppressing tumour antigen presentation.

Dual sgRNAs facilitate CRISPR/Cas9-mediated mouse genome targeting

TL;DR: It is reported that simultaneous use of dual sgRNAs to target an individual gene significantly improved the Cas9‐mediated genome targeting with a bi‐allelic modification efficiency of up to 78% and the target gene modifications were characterized by efficient germline transmission and site‐dependent off‐target effects.
