Generation of Gene-Modified Cynomolgus Monkey via Cas9/RNA-Mediated Gene Targeting in One-Cell Embryos
Yuyu Niu,Bin Shen,Yiqiang Cui,Yongchang Chen,Jianying Wang,Lei Wang,Yu Kang,Xiaoyang Zhao,Wei Si,Wei Li,Andy Peng Xiang,Jiankui Zhou,Xuejiang Guo,Ye Bi,Chenyang Si,Bian Hu,Guoying Dong,Hong Wang,Zuomin Zhou,Tianqing Li,Tao Tan,Xiuqiong Pu,Fang Wang,Shaohui Ji,Qi Zhou,Xingxu Huang,Weizhi Ji,Jiahao Sha +27 more
TL;DR: By coinjection of Cas9 mRNA and sgRNAs into one-cell-stage embryos, this system successfully achieves precise gene targeting in cynomolgus monkeys and enables simultaneous disruption of two target genes in one step, and no off-target mutagenesis was detected by comprehensive analysis.
Journal ArticleDOI
Ythdc2 is an N6-methyladenosine binding protein that regulates mammalian spermatogenesis.
Phillip J. Hsu,Yunfei Zhu,Honghui Ma,Honghui Ma,Yueshuai Guo,Xiaodan Shi,Yuanyuan Liu,Meijie Qi,Zhike Lu,Zhike Lu,Hailing Shi,Hailing Shi,Jianying Wang,Yiwei Cheng,Guan-Zheng Luo,Guan-Zheng Luo,Qing Dai,Qing Dai,Mingxi Liu,Xuejiang Guo,Jiahao Sha,Bin Shen,Chuan He,Chuan He +23 more
TL;DR: YTHDC2 is an m6A-binding protein that plays critical roles during spermatogenesis and enhances the translation efficiency of its targets and also decreases their mRNA abundance.

Anti-tumour immunity controlled through mRNA m 6 A methylation and YTHDF1 in dendritic cells
Dali Han,Dali Han,Jun Liu,Jun Liu,Chuanyuan Chen,Chuanyuan Chen,Lihui Dong,Yi Liu,Renbao Chang,Renbao Chang,Xiaona Huang,Yuanyuan Liu,Jianying Wang,Urszula Dougherty,Marc Bissonnette,Bin Shen,Ralph R. Weichselbaum,Meng Michelle Xu,Chuan He,Chuan He +19 more
TL;DR: It is shown that durable neoantigen-specific immunity is regulated by mRNA N6-methyadenosine (m6A) methylation through the m6A-binding protein YTHDF15, which suppresses the clearance of tumours by enhancing the translation of lysosomal proteases in dendritic cells and thereby suppressing tumour antigen presentation.
Journal ArticleDOI
Dual sgRNAs facilitate CRISPR/Cas9-mediated mouse genome targeting
Jiankui Zhou,Jianying Wang,Bin Shen,Li Chen,Yang Su,Jing Yang,Wensheng Zhang,Xuemei Tian,Xingxu Huang +8 more
TL;DR: It is reported that simultaneous use of dual sgRNAs to target an individual gene significantly improved the Cas9‐mediated genome targeting with a bi‐allelic modification efficiency of up to 78% and the target gene modifications were characterized by efficient germline transmission and site‐dependent off‐target effects.
