Efficient genome engineering in human pluripotent stem cells using Cas9 from Neisseria meningitidis

TL;DR: Efficient targeting of an endogenous gene in three hPSC lines using HDR is demonstrated using a distinct CRISPR-Cas system identified in Neisseria meningitidis, which is distinct from the commonly used Streptococcus pyogenes system.
Journal ArticleDOI

Human pluripotent stem cell-derived neural constructs for predicting neural toxicity

TL;DR: Stem cell biology, tissue engineering, bioinformatics, and machine learning were combined to implement an in vitro human cellular model for developmental neurotoxicity screening and demonstrated the value of human cell-based assays for predictive toxicology and should be useful for both drug and chemical safety assessment.
Journal ArticleDOI

Complement C3aR Inactivation Attenuates Tau Pathology and Reverses an Immune Network Deregulated in Tauopathy Models and Alzheimer’s Disease

TL;DR: It is shown that the expression of C3 and C3a receptor (C3aR1) are positively correlated with cognitive decline and Braak staging in human AD brains and a crucial role for activation of the C3-C3 aR network in mediating neuroinflammation and tau pathology is demonstrated.
