Human Leukocyte Antigen Class I-Restricted Activation of CD8+ T Cells Provides the Immunogenetic Basis of a Systemic Drug Hypersensitivity
Diana Chessman,Lyudmila Kostenko,Tessa Lethborg,Anthony W. Purcell,Nicholas A. Williamson,Zhenjun Chen,Lars Kjer-Nielsen,Nicole A. Mifsud,Brian D. Tait,Rhonda Holdsworth,Coral Ann Almeida,David Nolan,W.A. Macdonald,Julia K. Archbold,Anthony D Kellerher,Deborah Marriott,Simon Mallal,Mandvi Bharadwaj,Jamie Rossjohn,James McCluskey +19 more
TL;DR: It is shown that systemic reactions to abacavir were driven by drug-specific activation of cytokine-producing, cytotoxic CD8+ T cells, suggesting a basis for the strong HLA class I-association with certain inflammatory disorders.
Journal ArticleDOI
A naturally selected dimorphism within the HLA-B44 supertype alters class I structure, peptide repertoire, and T cell recognition.
W.A. Macdonald,Anthony W. Purcell,Nicole A. Mifsud,Lauren Kate Ely,David S. Williams,Linus Chang,Jeffrey J. Gorman,Craig Steven Clements,Lars Kjer-Nielsen,David M. Koelle,Scott R. Burrows,Brian D. Tait,Rhonda Holdsworth,Andrew G. Brooks,George O. Lovrecz,Louis Lu,Jamie Rossjohn,James McCluskey +17 more
TL;DR: The findings suggest that these closely related class I genes are maintained in diverse human populations through their differential impact on the selection of peptide ligands and the T cell repertoire.
Journal ArticleDOI
Human CD8+ T cell cross-reactivity across influenza A, B and C viruses
Marios Koutsakos,Patricia T. Illing,Thi H. O. Nguyen,Nicole A. Mifsud,Jeremy Chase Crawford,Simone Rizzetto,Auda A. Eltahla,E. Bridie Clemens,Sneha Sant,Brendon Y. Chua,Brendon Y. Thomas,Stephanie Gras,Stephanie Gras,Anthony W. Purcell,Katherine Kedzierska +48 more
TL;DR: This study introduces a new paradigm whereby CD8+ T cells confer unprecedented cross-reactivity across all influenza viruses, a key finding for the design of universal vaccines.
Journal ArticleDOI
Natural HLA class I polymorphism controls the pathway of antigen presentation and susceptibility to viral evasion.
D. Zernich,Anthony W. Purcell,W.A. Macdonald,Lars Kjer-Nielsen,Lauren Kate Ely,Nihay Laham,Tanya Crockford,Nicole A. Mifsud,Mandvi Bharadwaj,Linus Chang,Brian D. Tait,Rhonda Holdsworth,Andrew G. Brooks,Stephen P. Bottomley,Travis Clarke Beddoe,Chen Au Peh,Jamie Rossjohn,James McCluskey +17 more
TL;DR: A new layer of MHC polymorphism is unveiled that affects the generic pathway of Ag loading, revealing an unsuspected evolutionary trade-off in selection for optimal HLA class I loading versus effective pathogen evasion.
Journal ArticleDOI
Crystal structure of the human T cell receptor CD3εγ heterodimer complexed to the therapeutic mAb OKT3
Lars Kjer-Nielsen,Michelle A. Dunstone,Lyudmila Kostenko,Lauren Kate Ely,Travis Clarke Beddoe,Nicole A. Mifsud,Anthony W. Purcell,Andrew G. Brooks,James McCluskey,Jamie Rossjohn +9 more
TL;DR: The crystal structure of the human CD3eγ heterodimer is described to 2.1-A resolution complexed with OKT3, a therapeutic mAb that not only activates and tolerizes mature T cells but also induces regulatory T cells.
