Journal ArticleDOI
Broad host range of SARS-CoV-2 predicted by comparative and structural analysis of ACE2 in vertebrates.
Joana Damas,Graham M. Hughes,Kathleen C. Keough,Corrie A. Painter,Nicole S. Persky,Marco Corbo,Michael Hiller,Klaus-Peter Koepfli,Andreas R. Pfenning,Huabin Zhao,Huabin Zhao,Diane P. Genereux,Ross Swofford,Katherine S. Pollard,Oliver A. Ryder,Martin T. Nweeia,Martin T. Nweeia,Martin T. Nweeia,Kerstin Lindblad-Toh,Kerstin Lindblad-Toh,Emma C. Teeling,Elinor K. Karlsson,Elinor K. Karlsson,Harris A. Lewin +23 more
TL;DR: A large number of mammals that can potentially be infected by SARS-CoV-2 via their ACE2 proteins are identified to assist the identification of intermediate hosts for Sars-Cov-2 and hence reduce the opportunity for a future outbreak of COVID-19.
Journal ArticleDOI
Acquired HER2 mutations in ER+ metastatic breast cancer confer resistance to estrogen receptor-directed therapies.
Utthara Nayar,Ofir Cohen,Christian Kapstad,Christian Kapstad,Michael S. Cuoco,Adrienne G. Waks,Seth A. Wander,Corrie A. Painter,Samuel S. Freeman,Nicole S. Persky,Lori Marini,Karla Helvie,Nelly Oliver,Orit Rozenblatt-Rosen,Cynthia X. Ma,Aviv Regev,Aviv Regev,Eric P. Winer,Eric P. Winer,Nan Lin,Nan Lin,Nikhil Wagle +21 more
TL;DR: Activating HER2 mutations are shown to confer resistance to ER-directed therapies in patients with ER+ metastatic breast cancer, and these mutations conferred estrogen independence as well as resistance to tamoxifen, fulvestrant and the CDK4 and CDK6 inhibitor palbociclib.

TL;DR: A protein structural analysis was employed to qualitatively assess whether amino acid changes at variable residues would be likely to disrupt ACE2/SARS-CoV-2 binding, and found the number of predicted unfavorable changes significantly correlated with the binding score.
Journal ArticleDOI
Phenotypic Characterization of a Comprehensive Set of MAPK1/ERK2 Missense Mutants.
Lisa Brenan,Aleksandr Andreev,Ofir Cohen,Sasha Pantel,Atanas Kamburov,Davide Cacchiarelli,Nicole S. Persky,Cong Zhu,Mukta Bagul,Eva M. Goetz,Eva M. Goetz,Alex B. Burgin,Levi A. Garraway,Levi A. Garraway,Gad Getz,Gad Getz,Tarjei S. Mikkelsen,Federica Piccioni,David E. Root,Cory M. Johannessen +19 more
TL;DR: This work functionally characterized nearly all missense mutants of MAPK1/ERK2, an essential effector of oncogenic RAS and RAF, and discovered rare gain- and loss-of-function ERK2 mutants found in human tumors, revealing that, in the context of this assay, mutational frequency alone cannot identify all functionally impactful mutants.
Proceedings ArticleDOI
Abstract LB002: Mechanisms of acquired resistance to KRAS G12C inhibition in cancer
Mark M. Awad,Shengwu Liu,Kathryn C. Arbour,Viola W. Zhu,Melissa Lynne Johnson,Rebecca S. Heist,Tejas Patil,Gregory J. Riely,Joseph O. Jacobson,Julien Dilly,Xiaoping Yang,Nicole S. Persky,David E. Root,Lynette M. Sholl,Lee Lim,Kavita Garg,Mark Li,Lars D. Engstrom,Laura Waters,J. David Lawson,Peter D. Olson,James J. Christensen,Piro Lito,Sai-Hong Inatius Ou,Pasi A. Jänne,Andrew J. Aguirre +25 more
TL;DR: Diverse genomic and histologic mechanisms impart resistance to covalent KRASG12C inhibitors in patients with cancer, suggesting potential therapeutic strategies for overcoming drug resistance to specific mutations.
