Journal ArticleDOI
Complex Oscillatory Waves Emerging from Cortical Organoids Model Early Human Brain Network Development.
Cleber A. Trujillo,Richard Gao,Priscilla D. Negraes,Jing Gu,Justin Buchanan,Sebastian Preissl,Allen Wang,Wei Wu,Gabriel G. Haddad,Isaac A. Chaim,Alain Domissy,Matthieu Vandenberghe,Anna Devor,Gene W. Yeo,Bradley Voytek,Alysson R. Muotri +15 more
TL;DR: Human cortical organoids that dynamically change cellular populations during maturation and exhibited consistent increases in electrical activity over the span of several months show that the development of structured network activity in a human neocortex model may follow stable genetic programming.
Journal ArticleDOI
Modeling of TREX1-Dependent Autoimmune Disease using Human Stem Cells Highlights L1 Accumulation as a Source of Neuroinflammation.
Charles A. Thomas,Leon Tejwani,Cleber A. Trujillo,Priscilla D. Negraes,Roberto H. Herai,Pinar Mesci,Angela Macia,Yanick J. Crow,Alysson R. Muotri +8 more
TL;DR: In this model, reverse-transcriptase inhibitors rescued the neurotoxicity of AGS neurons and organoids, highlighting their potential utility in therapeutic regimens for AGS and related disorders.
Journal ArticleDOI
Zika Virus Targets Glioblastoma Stem Cells through a SOX2-Integrin αvβ5 Axis.

TL;DR: It is demonstrated that ZIKV preferentially infected and killed GSCs and stem-like cells in medulloblastoma and ependymoma in a SOX2-dependent manner, providing potential brain tumor therapy.
Journal ArticleDOI
Reintroduction of the archaic variant of NOVA1 in cortical organoids alters neurodevelopment.
Cleber A. Trujillo,Edward S. Rice,Nathan K. Schaefer,Isaac A. Chaim,Emily C. Wheeler,Assael A. Madrigal,Justin Buchanan,Sebastian Preissl,Allen Wang,Priscilla D. Negraes,Ryan A. Szeto,Roberto H. Lautz,Katerina Semendeferi,Beth Shapiro,Gene W. Yeo,Stephen E. P. Smith,Stephen E. P. Smith,Richard E. Green,Alysson R. Muotri +28 more
TL;DR: In this paper, the evolutionarily conserved splicing regulator neuro-oncological ventral antigen 1 (NOVA1) plays a key role in neural development and function.
Journal ArticleDOI
Chemical genetic identification of CDKL5 substrates reveals its role in neuronal microtubule dynamics.
Lucas L. Baltussen,Priscilla D. Negraes,Margaux Silvestre,Suzanne Claxton,Max Moeskops,Evangelos Christodoulou,Helen R. Flynn,Ambrosius P. Snijders,Alysson R. Muotri,Sila K. Ultanir +9 more
TL;DR: In CDKL5 knockout mouse neurons, dendritic microtubules have longer EB3‐labelled plus‐end growth duration and these altered dynamics are rescued by reduction of MAP1S levels through shRNA expression, indicating that CDKl5 regulates microtubule dynamics via phosphorylation ofMAP1S.
