The EUROclass trial: defining subgroups in common variable immunodeficiency
Claudia Wehr,Teemu Kivioja,Christian Schmitt,Berne Ferry,Torsten Witte,Efrem Eren,Marcela Vlková,M. Hernández,Drahomíra Detková,Philip R. Bos,Gonke Poerksen,Horst von Bernuth,Ulrich Baumann,Sigune Goldacker,Sylvia Gutenberger,Michael Schlesier,Florence Bergeron-van der Cruyssen,Magali Le Garff,Patrice Debré,Roland Jacobs,John R Jones,E. Bateman,Jiri Litzman,P. Martin van Hagen,Alessandro Plebani,Reinhold E. Schmidt,Vojtech Thon,Isabella Quinti,Teresa Espanol,A. David B. Webster,Helen Chapel,Mauno Vihinen,Eric Oksenhendler,Hans-Hartmut Peter,Klaus Warnatz +34 more
TL;DR: The clinical evaluation of 303 patients with the established diagnosis of CVID demonstrated a significant coincidence of granulomatous disease, autoimmune cytopenia, and splenomegaly, and an improved classification for CVID (EUROclass), separating patients with nearly absent B cells, and expansion of transitional or CD21(low) B cells.
Journal ArticleDOI
Circulating CD21low B cells in common variable immunodeficiency resemble tissue homing, innate-like B cells
Mirzokhid Rakhmanov,Baerbel Keller,Sylvia Gutenberger,Christian Foerster,Manfred Hoenig,Gertjan J. Driessen,Mirjam van der Burg,Jacques J.M. van Dongen,Elisabeth Wiech,Marcella Visentini,Isabella Quinti,Antje Prasse,Nadine Voelxen,Ulrich Salzer,Sigune Goldacker,Paul Fisch,Hermann Eibel,Klaus Schwarz,Hans-Hartmut Peter,Klaus Warnatz +19 more
TL;DR: It is suggested that CD21low B cells represent a human innate-like B cell population characterized by the low expression of CD21, which has close resemblance to the gene expression profile, phenotype, function and preferential tissue homing of murine B1 B cells.
Journal ArticleDOI
Multicenter experience in hematopoietic stem cell transplantation for serious complications of common variable immunodeficiency
Claudia Wehr,Andrew R. Gennery,Caroline A. Lindemans,Ansgar Schulz,Manfred Hoenig,Reinhard Marks,Mike Recher,Bernd Gruhn,Andreas Holbro,Ingmar Heijnen,Deborah Meyer,Goetz Ulrich Grigoleit,Hermann Einsele,Ulrich Baumann,Thorsten Witte,Karl Walter Sykora,Sigune Goldacker,Lorena Regairaz,Serap Aksoylar,Ömür Ardeniz,Marco Zecca,Przemysław Zdziarski,Isabelle Meyts,Susanne Matthes-Martin,Kohsuke Imai,Chikako Kamae,Adele K. Fielding,Suranjith L. Seneviratne,Nizar Mahlaoui,Mary Slatter,Tayfun Güngör,Peter D. Arkwright,Joris M. van Montfrans,Kathleen E. Sullivan,Bodo Grimbacher,Andrew J. Cant,Hans-Hartmut Peter,Juergen Finke,H. Bobby Gaspar,Klaus Warnatz,Marta Rizzi +40 more
TL;DR: HSCT in patients with CVID was beneficial in most surviving patients; however, there was a high mortality associated with the procedure, so this therapeutic approach should only be considered in carefully selected patients in whom there has been extensive characterization of the immunologic and/or genetic defect underlying the CVID diagnosis.
Journal ArticleDOI
Screening of 181 Patients With Antibody Deficiency for Deficiency of Adenosine Deaminase 2 Sheds New Light on the Disease in Adulthood
Johanna Schepp,Michele Proietti,Natalie Frede,Mary Buchta,Katrin Hübscher,Jessica Rojas Restrepo,Sigune Goldacker,Klaus Warnatz,Jana Pachlopnik Schmid,Andrea Duppenthaler,Vassilios Lougaris,Ignacio Uriarte,Susan J. Kelly,Michael S. Hershfield,Bodo Grimbacher +14 more
TL;DR: It is shown that deficiency of adenosine deaminase 2 (DADA2) in patients with antibody deficiency and the clinical picture of the disease in adulthood in adulthood is similar to that of absence of DADA2.
Journal ArticleDOI
The TH1 phenotype of follicular helper T cells indicates an IFN-γ-associated immune dysregulation in patients with CD21low common variable immunodeficiency.
Susanne Unger,Maximilian Seidl,Maximilian Seidl,Pauline A. van Schouwenburg,Mirzokhid Rakhmanov,Alla Bulashevska,Natalie Frede,Bodo Grimbacher,Jens Pfeiffer,Klaudia Schrenk,Klaudia Schrenk,Luis E. Muñoz,Leif G. Hanitsch,Ina Stumpf,Fabian M.P. Kaiser,Oliver Hausmann,Florian Kollert,Sigune Goldacker,Mirjam van der Burg,Baerbel Keller,Klaus Warnatz +20 more
TL;DR: Identification of excessive IFN‐&ggr; production by blood and lymph node–derived T cells of patients with CVID with immune dysregulation will offer new therapeutic avenues for this subgroup.
