Journal ArticleDOI
Genome-wide CRISPR screen identifies TMEM41B as a gene required for autophagosome formation.
Keigo Morita,Yutaro Hama,Tamaki Izume,Norito Tamura,Toshihide Ueno,Yoshihiro Yamashita,Yuriko Sakamaki,Kaito Mimura,Hideaki Morishita,Wataru Shihoya,Osamu Nureki,Hiroyuki Mano,Noboru Mizushima +12 more
TL;DR: Results suggest that TMEM41B and VMP1 function together at an early step of autophagosome formation, and overexpression of V MP1 restored autophagic flux in TMEM 41B-KO cells.
Journal ArticleDOI
X-ray structures of endothelin ETB receptor bound to clinical antagonist bosentan and its analog
Wataru Shihoya,Wataru Shihoya,Tomohiro Nishizawa,Tomohiro Nishizawa,Keitaro Yamashita,Asuka Inoue,Asuka Inoue,Kunio Hirata,Francois Marie Ngako Kadji,Akiko Okuta,Kazutoshi Tani,Junken Aoki,Junken Aoki,Yoshinori Fujiyoshi,Tomoko Doi,Osamu Nureki +15 more
TL;DR: Crystal structures of human endothelin ETB receptor bound to bosentan and to the ETB-selective analog K-8794 reveal the detailed water-mediated hydrogen-bonding network at the transmembrane core and reveal detailed interactions with ETB, which is probably conserved in the ETA receptor.
Journal ArticleDOI
Crystal structure of heliorhodopsin.
Wataru Shihoya,Keiichi Inoue,Manish Singh,Masae Konno,Shoko Hososhima,Keitaro Yamashita,Kento Ikeda,Akimitsu Higuchi,Tamaki Izume,Sae Okazaki,Masanori Hashimoto,Ritsu Mizutori,Sahoko Tomida,Yumeka Yamauchi,Rei Abe-Yoshizumi,Kota Katayama,Satoshi P. Tsunoda,Satoshi P. Tsunoda,Mikihiro Shibata,Yuji Furutani,Yuji Furutani,Yuji Furutani,Alina Pushkarev,Oded Béjà,Takayuki Uchihashi,Takayuki Uchihashi,Hideki Kandori,Osamu Nureki +27 more
TL;DR: The 2.4-Å-resolution structure of HeR from an uncultured Thermoplasmatales archaeon SG8-52-1 shows that it adopts a similar fold to that of type I rhodopsin—despite the low sequence identity—but there are also several marked differences that provide insights into heliorhodopin function.
Journal ArticleDOI
Crystal structures of human ET B receptor provide mechanistic insight into receptor activation and partial activation
Wataru Shihoya,Tamaki Izume,Asuka Inoue,Keitaro Yamashita,Francois Marie Ngako Kadji,Kunio Hirata,Junken Aoki,Junken Aoki,Tomohiro Nishizawa,Osamu Nureki +9 more
TL;DR: The current findings clarify the detailed molecular mechanism for the coupling between the orthosteric pocket and the G-protein binding, and the partial agonistic effect of IRL1620, thus paving the way for the design of improved agonistic drugs targeting ETB.
Journal ArticleDOI
Time-resolved serial femtosecond crystallography reveals early structural changes in channelrhodopsin
Kazumasa Oda,Takashi Nomura,Takanori Nakane,Keitaro Yamashita,Keiichi Inoue,Shota Ito,Johannes Vierock,Kunio Hirata,Andrés D. Maturana,Kota Katayama,Tatsuya Ikuta,Itsuki Ishigami,Tamaki Izume,Rie Umeda,Ryuun Eguma,Satomi Oishi,Go Kasuya,Takafumi Kato,Tsukasa Kusakizako,Wataru Shihoya,Hiroto Shimada,Tomoyuki Takatsuji,Mizuki Takemoto,Reiya Taniguchi,Atsuhiro Tomita,Ryoki Nakamura,Masahiro Fukuda,Hirotake Miyauchi,Yongchan Lee,Eriko Nango,Rie Tanaka,Tomoyuki Tanaka,Michihiro Sugahara,Tetsunari Kimura,Tatsuro Shimamura,Takaaki Fujiwara,Yasuaki Yamanaka,Shigeki Owada,Yasumasa Joti,Kensuke Tono,Ryuichiro Ishitani,Shigehiko Hayashi,Hideki Kandori,Peter Hegemann,So Iwata,Minoru Kubo,Tomohiro Nishizawa,Tomohiro Nishizawa,Osamu Nureki +48 more
TL;DR: In this article, the authors performed time-resolved serial femtosecond crystallographic analyses of ChRs by using an X-ray free electron laser, which revealed conformational changes following photoactivation.
