Abstract: Introduction Numerous studies have examined the validity of accelerometry-based activity monitors but few studies have systematically studied the reliability of different accelerometer units for assessing a standardized bout of physical activity. Improving understanding of error in these devices is an important research objective because they are increasingly being used in large surveillance studies and intervention trials that require the use of multiple units over time. Methods Four samples of college-aged participants were recruited to collect reliability data on four different accelerometer types (CSA/MTI, Biotrainer Pro, Tritrac-R3D, and Actical). The participants completed three trials of treadmill walking (3 mph) while wearing multiple units of a specific monitor type. For each trial, the participant completed a series of 5-min bouts of walking (one for each monitoring unit) with 1-min of standing rest between each bout. Generalizability (G) theory was used to quantify variance components associated with individual monitor units, trials, and subjects as well as interactions between these terms. Results The overall G coefficients range from 0.43 to 0.64 for the four monitor types. Corresponding intraclass correlation coefficients (ICC) ranged from 0.62 to 0.80. The CSA/MTI was found to have the least variability across monitor units and trials and the highest overall reliability. The Actical was found to have the poorest reliability. Conclusion The CSA/MTI appeared to have acceptable reliability for most research applications (G values above 0.60 and ICC values above 0.80), but values with the other devices indicate some possible concerns with reliability. Additional work is needed to better understand factors contributing to variability in accelerometry data and to determine appropriate calibration protocols to improve reliability of these measures for different research applications.

Abstract: The reconstruction of the signal from hadrons and jets emerging from the proton–proton collisions at the Large Hadron Collider (LHC) and entering the ATLAS calorimeters is based on a three-dimensional topological clustering of individual calorimeter cell signals. The cluster formation follows cell signal-significance patterns generated by electromagnetic and hadronic showers. In this, the clustering algorithm implicitly performs a topological noise suppression by removing cells with insignificant signals which are not in close proximity to cells with significant signals. The resulting topological cell clusters have shape and location information, which is exploited to apply a local energy calibration and corrections depending on the nature of the cluster. Topological cell clustering is established as a well-performing calorimeter signal definition for jet and missing transverse momentum reconstruction in ATLAS.

Abstract: The ability to precisely engineer plant genomes offers much potential for advancing basic and applied plant biology. Here, we describe methods for the targeted modification of plant genomes using transcription activator-like effector nucleases (TALENs). Methods were optimized using tobacco (Nicotiana tabacum) protoplasts and TALENs targeting the acetolactate synthase (ALS) gene. Optimal TALEN scaffolds were identified using a protoplast-based single-strand annealing assay in which TALEN cleavage creates a functional yellow fluorescent protein gene, enabling quantification of TALEN activity by flow cytometry. Single-strand annealing activity data for TALENs with different scaffolds correlated highly with their activity at endogenous targets, as measured by high-throughput DNA sequencing of polymerase chain reaction products encompassing the TALEN recognition sites. TALENs introduced targeted mutations in ALS in 30% of transformed cells, and the frequencies of targeted gene insertion approximated 14%. These efficiencies made it possible to recover genome modifications without selection or enrichment regimes: 32% of tobacco calli generated from protoplasts transformed with TALEN-encoding constructs had TALEN-induced mutations in ALS, and of 16 calli characterized in detail, all had mutations in one allele each of the duplicate ALS genes (SurA and SurB). In calli derived from cells treated with a TALEN and a 322-bp donor molecule differing by 6 bp from the ALS coding sequence, 4% showed evidence of targeted gene replacement. Abstract: Five experiments examined effects of songs with violent lyrics on aggressive thoughts and hostile feelings. Experiments 1, 3, 4 and 5 demonstrated that college students who heard a violent song felt more hostile than those who heard a similar but nonviolent song. Experiments 2-5 demonstrated a similar increase in aggressive thoughts. These effects replicated across songs and song types (e.g., rock, humorous, nonhumorous). Experiments 3-5 also demonstrated that trait hostility was positively related to state hostility but did not moderate the song lyric effects. Discussion centers on the potential role of lyric content on aggression in short-term settings, relation to catharsis and other media violence domains, development of aggressive personality, differences between long-term and short-term effects, and possible mitigating factors.
