Relationship of δ-Aminolevulinic Acid-Induced Protoporphyrin IX Levels to Mitochondrial Content in Neoplastic Cells in Vitro

TLDR
The data demonstrate that enzymes involved in heme biosynthesis, in general, display a rank order associated with mitochondrial content, and imply that such parameters may have value as prognosticators of cells to produce delta-aminolevulinic acid-induced protoporphyrin IX, a photosensitizer for photodynamic therapy of cancer.

Milestones in the development of photodynamic therapy and fluorescence diagnosis

TL;DR: Basic aspects of PDT will be focused on, all important for the route of development leading PDT to its present state: early work on hematoporphyrin and hematocarbon derivative, second and third generation photosensitizers, 5-aminolevulinic acid and its derivatives, oxygen and singlet oxygen.

A trial of short incubation, broad-area photodynamic therapy for facial actinic keratoses and diffuse photodamage.

TL;DR: This delta-ALA/PDT protocol is safe and effective for AK treatment as well as for improving photodamage and further studies with a larger cohort, longer follow-up, and histologic confirmation of the clinical data are of value.

Review of Neurosurgical Fluorescence Imaging Methodologies

TL;DR: Currently, optimized probe approaches are showing efficacy in clinical trials, and fully commercialized imaging systems are emerging, which will clearly help to lead adoption into neurosurgical practice.

Metabolic characterization of tumor cell-specific protoporphyrin IX accumulation after exposure to 5-aminolevulinic acid in human colonic cells.

TL;DR: Further research of tumor cell selectivity of PPIX fluorescence should focus on the mechanisms responsible for an altered PPIX metabolism to find tumor-specific target molecules, thus leading to an improved clinical practicability of ALA application and consequent endoscopy.

Rapid colorimetric assay for cellular growth and survival: Application to proliferation and cytotoxicity assays

TL;DR: A tetrazolium salt has been used to develop a quantitative colorimetric assay for mammalian cell survival and proliferation and is used to measure proliferative lymphokines, mitogen stimulations and complement-mediated lysis.

Characterization of the cellular reduction of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT): subcellular localization, substrate dependence, and involvement of mitochondrial electron transport in MTT reduction

TL;DR: Most cellular reduction of MTT occurs extramitochondrially and probably involves the pyridine nucleotide cofactors NADH and NADPH-dependent mechanisms that are insensitive to respiratory chain inhibitors.

Determination of [3h]- and [14c]hematoporphyrin derivative distribution in malignant and normal tissue.

TL;DR: The results tend to disprove the generalization that HPD accumulates in malignant tissue to a higher degree than in all normal tissue and it is reported that gross visualization of porphyrin fluorescence cannot be correlated with actual tissue concentrations of the dye.
