Structure-function relationship in allosteric aspartate carbamoyltransferase from Escherichia coli. I. Primary structure of a pyrI gene encoding a modified regulatory subunit.

TLDR
The sequence of the mutated pyrI gene is reported and it is shown that, during the generation of this pyrBI-bearing phage, six codons from lambda DNA have been substituted for the eight terminal codons of the wild-type gene.

Allosteric Regulation of Catalytic Activity:Escherichia coli Aspartate Transcarbamoylase versus Yeast Chorismate Mutase

TL;DR: This review compares the principles of allosteric transitions of the complex classical model aspartate transcarbamoylase from Escherichia coli and the less complicated chorismate mutase derived from baker's yeast, which functions as a homodimer.
Journal ArticleDOI

Crystal structure of CTP-ligated T state aspartate transcarbamoylase at 2.5 A resolution: implications for ATCase mutants and the mechanism of negative cooperativity.

TL;DR: The location of the first seven residues of the Regulatory chain has been identified for the first time in a refined ATCase crystal structure, and the proximity of this portion of the regulatory chain to the allosteric site suggests a potential role for these residues in nucleotide binding to the enzyme.
Journal ArticleDOI

Molecular evolution and genetic engineering of protein domains involving aspartate transcarbamoylase.

TL;DR: A comparison of the ATCase and OTCases with respect to Regulatory Control in Bacteria and Conservation of Important Amino Acid Residues in Catalysis and Regulation is presented.

Sequencing end-labeled DNA with base-specific chemical cleavages.

TL;DR: The chapter presents techniques for producing discrete DNA fragments, end-labeling DNA, segregating end- labeled fragments, extracting DNA from gels, and the protocols for partially cleaving it at specific bases using the chemical reactions.

On the Nature of Allosteric Transitions: A Plausible Model

TL;DR: "It is certain that all bodies whatsoever, though they have no sense, yet they have perception, and whether the body be alterant or alterec, evermore a perception precedeth operation; for else all bodies would be like one to another."
Journal ArticleDOI

Calcium-dependent bacteriophage DNA infection.

TL;DR: Escherichia coli cells of strain K12 and C can be made competent to take up temperate phage DNA without the use of “helper phage”, and is effective for both linear and circular DNA molecules.

A new pair of M13 vectors for selecting either DNA strand of double-digest restriction fragments.

TL;DR: Two new M13 vectors, M13MP8 and M13mp9, have been constructed that permit the cloning of the same restriction fragment in both possible orientations, allowing the use of only one of the two DNA strands as a template for M13 shotgun sequencing.
