DNA Demethylation in Zebrafish Involves the Coupling of a Deaminase, a Glycosylase, and Gadd45
TL;DR: Using zebrafish embryos, evidence is provided for a coupled mechanism of 5-meC demethylation, whereby AID deaminates 5- meC, followed by thymine base excision by Mbd4, promoted by Gadd45.

TL;DR: It is shown that homozygous loss of APC causes failed intestinal cell differentiation but that this occurs in the absence of nuclear beta-catenin and increased intestinal cell proliferation, which suggests that, following APC loss, CtBP1 contributes to adenoma initiation as a first step, whereas KRAS activation and beta-Catenin nuclear localization promote adenomas progression to carcinomas as a second step.
