A Modified γ-Retrovirus Vector for X-Linked Severe Combined Immunodeficiency
Salima Hacein-Bey-Abina,Sung-Yun Pai,H. Bobby Gaspar,Myriam Armant,Charles C. Berry,Stéphane Blanche,Jack J. Bleesing,Johanna Blondeau,Helen de Boer,Karen F. Buckland,Laure Caccavelli,Guilhem Cros,Satiro N. De Oliveira,Karen S. Fernandez,Dongjing Guo,Chad E. Harris,Gregory Hopkins,Leslie Lehmann,Annick Lim,Wendy B. London,Johannes C.M. Silberstein,Emmanuelle Six,Fabien Touzot,Alla V. Tsytsykova,Jinhua Xu-Bayford,Christopher Baum,Frederic D. Bushman,Alain Fischer,Donald B. Kohn,Alexandra H. Filipovich,Luigi D. Notarangelo,Marina Cavazzana,David A. Williams,Adrian J. Thrasher +48 more
TL;DR: This modified γ-retrovirus vector was found to retain efficacy in the treatment of SCID-X1 and the long-term effect of this therapy on leukemogenesis remains unknown.
Journal ArticleDOI
A primary T-cell immunodeficiency associated with defective transmembrane calcium influx.
F Le Deist,Claire Hivroz,M Partiseti,Caroline Thomas,Buc Ha,Matias Oleastro,B Belohradsky,Daniel Choquet,Alain Fischer +8 more
TL;DR: This primary T-cell immunodeficiency appears to be due to defective Ca2+ entry through the plasma membrane, which did not alter B-cell proliferation, platelet function, and polymorphonuclear neutrophil (PMN) function.
Journal ArticleDOI
Lymphoproliferative syndrome with autoimmunity: A possible genetic basis for dominant expression of the clinical manifestations.
Frédéric Rieux-Laucat,S Blachère,S Danielan,J P de Villartay,Matias Oleastro,Eric Solary,Brigitte Bader-Meunier,Peter D. Arkwright,C. Pondaré,Françoise Bernaudin,Helen Chapel,Susan Nielsen,M Berrah,A Fischer,F Le Deist +14 more
TL;DR: It was found that lymphoproliferative manifestations resolved with age, whereas immunological disorders persisted, suggesting that Fas-mediated apoptosis plays a more important role in lymphocyte homeostasis in early childhood than later on in life.
Journal ArticleDOI
Incomplete penetrance for isolated congenital asplenia in humans with mutations in translated and untranslated RPSA exons
Alexandre Bolze,Bertrand Boisson,Bertrand Boisson,Bertrand Boisson,Barbara Bosch,Alexander Antipenko,Matthieu Bouaziz,Matthieu Bouaziz,Paul Sackstein,Malik Chaker-Margot,Vincent Barlogis,Tracy A Briggs,Elena Colino,Aurora C. TL;DR: 11 new heterozygous ICA-causing RPSA protein-coding mutations, and the first two mutations in the 5′-UTR of this gene, which disrupt mRNA splicing are described, which can underlie ICA with complete or incomplete penetrance.
Journal ArticleDOI
Immune Reconstitution and Preliminary Safety Analysis Of 9 Patients Treated With Somatic Gene Therapy For X-Linked Severe Combined Immunodeficiency (SCID-X1) With a Self-Inactivating Gammaretroviral Vector
Salima Hacein-Bey-Abina,Frederic D. Bushman,Myriam Armant,Stéphane Blanche,Johanna Blondeau,Laure Caccavelli,Heather Daley,Colleen Dansereau,Satiro deOliveira,Dongjing Guo,Gregory Hopkins,Annick Lim,M.Sci,Wendy B. Van der Loo,Christopher Baum,H. Bobby Gaspar,Donald B. Kohn,Alexandra H. Filipovich,Luigi D. Notarangelo,Alain Fischer,Marina Cavazzana-Calvo,David A. Williams,Adrian J. Thrasher +34 more
TL;DR: The early immune recovery and peripheral blood insertion site analysis between 6 evaluable subjects on Trial 2 with the 20 subjects enrolled on the previous trials of MLV-based γc vector was compared and showed a lack of clonal skewing in primary and secondary transplant recipient mice in vivo and lack of insertions in either Lmo2 or Evi1 in these mice.
