Journal ArticleDOI
The epigenetics of aging and neurodegeneration.
Roy Lardenoije,Artemis Iatrou,Gunter Kenis,Konstantinos Kompotis,Harry W.M. Steinbusch,Diego Mastroeni,Paul D. Coleman,Cynthia A. Lemere,Patrick R. Hof,Daniel L.A. van den Hove,Bart P. F. Rutten +10 more
TL;DR: Current knowledge about the major epigenetic mechanisms, including DNA methylation and DNA demethylation, chromatin remodeling and non-coding RNAs, as well as the involvement of these mechanisms in normal aging and in the pathophysiology of the most common neurodegenerative diseases are reviewed.
Journal ArticleDOI
Epigenetically regulated microRNAs in Alzheimer's disease
Daniel L.A. van den Hove,Daniel L.A. van den Hove,Konstantinos Kompotis,Roy Lardenoije,Gunter Kenis,Jonathan Mill,Jonathan Mill,Harry W.M. Steinbusch,Klaus-Peter Lesch,Klaus-Peter Lesch,Carlos P. Fitzsimons,Bart De Strooper,Bart P. F. Rutten +12 more
TL;DR: This work reviews those miRNAs implicated in AD that are regulated by promoter DNA methylation and/or chromatin modifications and concludes with the delineation of a complex epigenetic-miRNA regulatory network and its alterations in AD.
Journal ArticleDOI
Alzheimer’s disease-associated (hydroxy)methylomic changes in the brain and blood
Roy Lardenoije,Roy Lardenoije,Janou A. Y. Roubroeks,Janou A. Y. Roubroeks,Ehsan Pishva,Ehsan Pishva,Markus Leber,Holger Wagner,Artemis Iatrou,Adam Smith,Rebecca G. Smith,Lars M. T. Coleman,Jonathan Mill,Jonathan Mill,Bart P. F. Rutten,Katie Lunnon,Alfredo Ramirez,Alfredo Ramirez,Daniel L.A. van den Hove,Daniel L.A. van den Hove +42 more
TL;DR: The implication of genome-wide significant differential methylation of OXT, encoding oxytocin, in two independent cohorts indicates it is a promising target for future studies on early biomarkers and novel therapeutic strategies in AD.
Journal ArticleDOI
Epigenetic regulation of adult neural stem cells: implications for Alzheimer's disease.
Carlos P. Fitzsimons,Emma J. van Bodegraven,Marijn Schouten,Roy Lardenoije,Konstantinos Kompotis,Gunter Kenis,Mark van den Hurk,Marco P. Boks,Caroline Biojone,Samia R. L. Joca,Harry W.M. Steinbusch,Katie Lunnon,Diego Mastroeni,Jonathan Mill,Paul J. Lucassen,Paul D. Coleman,Daniel L.A. van den Hove,Bart P. F. Rutten,Bart P. F. Rutten +18 more
TL;DR: The epigenetic mechanisms relevant for NSCs and AD etiology are reviewed, and the role of stem cells e.g. in hippocampal functions like cognition, and given their potential for brain repair, are reviewed.
Journal ArticleDOI
Age-related epigenetic changes in hippocampal subregions of four animal models of Alzheimer's disease.
Roy Lardenoije,Daniel L.A. van den Hove,Monique Havermans,Anne van Casteren,Kevin X. Le,Roberta M. Palmour,Cynthia A. Lemere,Bart P. F. Rutten +7 more
TL;DR: Only the J20 model showed an age‐related reduction in global DNA methylation, while DNA hypermethylation was observed in the 3xTg‐AD model, and differences between models suggest interaction genotype and epigenetic state.
