Journal ArticleDOI
Distinct external signals trigger sequential release of apical organelles during erythrocyte invasion by malaria parasites.
TL;DR: The results identify for the first time the external signals responsible for the sequential release of microneme and rhoptry proteins during erythrocyte invasion and provide a starting point for the dissection of signal transduction pathways involved in regulated exocytosis of these key apical organelles.
Journal ArticleDOI
Phosphoproteomics reveals malaria parasite Protein Kinase G as a signalling hub regulating egress and invasion
Mahmood M. Alam,Lev Solyakov,Andrew R. Bottrill,Christian Flueck,Faiza Amber Siddiqui,Shailja Singh,Sharad Mistry,Maria Viskaduraki,Kate D. Lee,Christine S. Hopp,Chetan E. Chitnis,Christian Doerig,Robert W. Moon,Judith L. Green,Anthony A. Holder,David A. Baker,Andrew B. Tobin +16 more
TL;DR: The phosphorylation sites on 69 proteins involved in cell signalling, proteolysis, gene regulation, protein export and ion and protein transport are identified, indicating that cGMP/PfPKG acts as a signalling hub that plays a central role in a number of core parasite processes.
Journal ArticleDOI
Perforin‐like protein PPLP2 permeabilizes the red blood cell membrane during egress of Plasmodium falciparum gametocytes
Christine C. Wirth,Svetlana Glushakova,Matthias Scheuermayer,Urska Repnik,Swati Garg,Dominik Schaack,Marika M. Kachman,Tim Weißbach,Joshua Zimmerberg,Thomas Dandekar,Gareth Griffiths,Chetan E. Chitnis,Shailja Singh,Shailja Singh,Rainer Fischer,Rainer Fischer,Gabriele Pradel,Gabriele Pradel +17 more
TL;DR: It is proposed that the haemolytic activity of PPLP2 is essential for gametocyte egress due to permeabilization of the erythrocyte membrane and depletion of the ______ cytoplasm.
Journal ArticleDOI
Calcium-dependent permeabilization of erythrocytes by a perforin-like protein during egress of malaria parasites.
Swati Garg,Shalini Agarwal,Saravanan Kumar,Syed Shams Yazdani,Chetan E. Chitnis,Shailja Singh,Shailja Singh +6 more
TL;DR: It is demonstrated that calcium-dependent permeabilization of host red blood cells is critical for egress of Plasmodium falciparum merozoites.
Journal ArticleDOI
Characterization of Plasmodium falciparum Calcium-dependent Protein Kinase 1 (PfCDPK1) and Its Role in Microneme Secretion during Erythrocyte Invasion
Abhisheka Bansal,Shailja Singh,Kunal R. More,Dhiraj Hans,Kuldeep Nangalia,Manickam Yogavel,Amit Sharma,Chetan E. Chitnis +7 more
TL;DR: In this article, a peptide P3 from the junction domain of Plasmodium falciparum CDPK1 (PfCDPK) as well as purfalcamine inhibit functional activity to block microneme discharge and erythrocyte invasion.
