KRASG12C Inhibition with Sotorasib in Advanced Solid Tumors.
David S. Hong,Marwan Fakih,John H. Strickler,Jayesh Desai,G. Durm,Geoffrey I. Shapiro,Gerald S. Falchook,Timothy J. Price,Adrian G. Sacher,Crystal S. Denlinger,Yung-Jue Bang,Grace K. Dy,John C. Krauss,Yasutoshi Kuboki,James Kuo,Andrew L. Coveler,Keunchil Park,Tae Won Kim,Fabrice Barlesi,Pamela N. Munster,Suresh S. Ramalingam,Timothy F. Burns,Funda Meric-Bernstam,H. Henary,J. Ngang,Gataree Ngarmchamnanrith,J. Kim,Brett E. Houk,Jude Canon,J. Russell Lipford,Gregory Friberg,Piro Lito,Ramaswamy Govindan,Bob T. Li +33 more
TL;DR: Sotorasib showed encouraging anticancer activity in patients with heavily pretreated advanced solid tumors harboring the KRAS p.G12C mutation and responded to pharmacokinetics and objective response according to Response Evaluation Criteria in Solid Tumors (RECIST), version 1.1.
Journal ArticleDOI
Targeting Heat Shock Proteins in Cancer: A Promising Therapeutic Approach
TL;DR: The roles of some of the important HSPs in cancer are reviewed, and how targeting them could be efficacious, especially when traditional cancer therapies fail are reviewed.
Journal ArticleDOI
Detection of cancer DNA in plasma of patients with early-stage breast cancer.
Julia A. Beaver,Danijela Jelovac,Sasidharan Balukrishna,Rory L. Cochran,Sarah Croessmann,Daniel J. Zabransky,Hong Yuen Wong,Patricia Valda Toro,Justin Cidado,Brian G. Blair,David Chu,Timothy F. Burns,Michaela J. Higgins,Vered Stearns,Lisa K. Jacobs,Mehran Habibi,Julie R. Lange,Paula J. Hurley,Josh Lauring,Dustin A. VanDenBerg,Jill Kessler,Stacie Jeter,Michael L. Samuels,Dianna Maar,Leslie Cope,Ashley Cimino-Mathews,Pedram Argani,Antonio C. Wolff,Ben Ho Park +28 more
TL;DR: This prospective study demonstrates accurate mutation detection in tumor tissues using ddPCR, and that ptDNA can be detected in blood before and after surgery in patients with early-stage breast cancer.
Journal ArticleDOI
Targeting the EMT transcription factor TWIST1 overcomes resistance to EGFR inhibitors in EGFR-mutant non-small-cell lung cancer.
Zachary A. Yochum,Jessica Cades,Hailun Wang,Suman Chatterjee,Brian W. Simons,James P. O’Brien,Susheel K. Khetarpal,Ghali Lemtiri-Chlieh,Kayla V. Myers,Eric H.-B. Huang,Charles M. Rudin,Phuoc T. Tran,Timothy F. Burns +12 more
TL;DR: TWIST1 is established as a driver of resistance to EGFR TKIs and rationale for use of TWIST1 inhibitors or BCL2 inhibitors as means to overcome EMT-mediated resistance toEGFR TkIs is provided.
Journal ArticleDOI
Twist1 suppresses senescence programs and thereby accelerates and maintains mutant Kras-induced lung tumorigenesis.
Phuoc T. Tran,Emelyn H. Shroff,Timothy F. Burns,Saravanan Thiyagarajan,Sandhya Das,Tahera Zabuawala,Joy Chen,Yoon Jae Cho,Richard Luong,Pablo Tamayo,T. Salih,Khaled Aziz,Stacey J. Adam,Silvestre Vicent,Carsten H. Nielsen,Nadia Withofs,Alejandro Sweet-Cordero,Sanjiv S. Gambhir,Charles M. Rudin,Dean W. Felsher +19 more
TL;DR: It is shown that Twist1 cooperates with KrasG12D to markedly accelerate lung tumorigenesis by abrogating cellular senescence programs and promoting the progression from benign adenomas to adenocarcinomas and the suppression of TWIST1 in human tumors may be an effective example of pro-senescence therapy.
