TL;DR: This study presents the first lysine-acetylation and lysin-succinylation proteome analysis of seedling leaves in Brachypodium distachyon L (Bd), and identifies two new acetylated motifs (K---K and K-I-K) and twelve significantly enriched succinylated motifS for the first time, which could serve as possible binding loci for future studies in plants.
Abstract: Protein acetylation and succinylation are the most crucial protein post-translational modifications (PTMs) involved in the regulation of plant growth and development. In this study, we present the first lysine-acetylation and lysine-succinylation proteome analysis of seedling leaves in Brachypodium distachyon L (Bd). Using high accuracy nano LC-MS/MS combined with affinity purification, we identified a total of 636 lysine-acetylated sites in 353 proteins and 605 lysine-succinylated sites in 262 proteins. These proteins participated in many biology processes, with various molecular functions. In particular, 119 proteins and 115 sites were found to be both acetylated and succinylated, simultaneously. Among the 353 acetylated proteins, 148 had acetylation orthologs in Oryza sativa L., Arabidopsis thaliana, Synechocystis sp. PCC 6803, and Glycine max L. Among the 262 succinylated proteins, 170 of them were found to have homologous proteins in Oryza sativa L., Escherichia coli, Sacchayromyces cerevisiae, or Homo sapiens. Motif-X analysis of the acetylated and succinylated sites identified two new acetylated motifs (K---K and K-I-K) and twelve significantly enriched succinylated motifs for the first time, which could serve as possible binding loci for future studies in plants. Our comprehensive dataset provides a promising starting point for further functional analysis of acetylation and succinylation in Bd and other plant species.

TL;DR: Correlation analysis and principal component suggested that Ni, Cu, V, Zn, Mn, Cr, Ti, and Pb were derived from soil erosion in upper reaches of the reservoir, while Fe, Cd, Hg, As, and partial Pb originated from anthropogenic sources, particularly industrial mining and gold tailings.
Abstract: Heavy metals concentrations in surface sediments from Miyun Reservoir were determined to evaluate the pollution and identify the sources. The average content of metals in sediments from Miyun Reservoir followed the order Al>Fe>Ti>Mn>V>Zn>Cr>Ni>Cu>Pb>As>Cd>Hg, and the most mean values were lower than the globe average shale. Heavy metals concentrations at the inflow area of Baihe were higher than those at the inflow area of Chaohe. Heavy metals pollution assessment was carried out by factor enrichment (EF), geoaccumulation index (I(geo)), and potential ecological risk (RI). The EF values for all heavy metals except Hg, Cd, and Cr at several sites were lower than 3, suggesting low anthropogenic impact on the metals level. The I(geo) values of Pb indicated that half of the sites were unpolluted to moderately polluted and mainly located in the Baihe area of the reservoir. The RI showed that heavy metals of Miyun Reservoir were low potential risk, however, Hg approached or belonged to moderate ecological risk at sites of M5, M7, and M13. Correlation analysis and principal component suggested that Ni, Cu, V, Zn, Mn, Cr, Ti, and Pb were derived from soil erosion in upper reaches of the reservoir, while Fe, Cd, Hg, As, and partial Pb originated from anthropogenic sources, particularly industrial mining and gold tailings.

TL;DR: In this paper, the generalized Riemann problem method (GRP) is used to evaluate the midpoint values of solutions at each cell interface so that the bottom topography effect is included in numerical fluxes, and at the same step the source term is discretized with an interface method in which only mid-point values are plugged in.
Abstract: This paper extends the generalized Riemann problem method (GRP) to the system of shallow water equations with bottom topography. The main contribution is that the generalized Riemann problem method (J. Comput. Phys. 1984; 55(1):1–32) is used to evaluate the midpoint values of solutions at each cell interface so that the bottom topography effect is included in numerical fluxes, and at the same step the source term is discretized with an interface method in which only mid-point values are plugged in. This scheme is well balanced between the flux gradient and bottom topography when incorporating the surface gradient method (SGM) (J. Comput. Phys. 2001; 168(1):1–25) into data reconstruction step, and it is also suitable for both steady and unsteady flow simulations. We illustrate the accuracy of this scheme by several 1-D and 2-D numerical experiments. Copyright © 2005 John Wiley & Sons, Ltd.
