TLDR
To identify gene products that function stoichiometrically in mitotic chromosome transmission, genes were cloned on high copy number plasmids and transformed into yeast cells, and the transformants were examined for an increase in the frequency of mitotic chromosomes loss or recombination resulting from the gene imbalance.

mRNA stability in mammalian cells.

TL;DR: This review concerns how cytoplasmic mRNA half-lives are regulated and how mRNA decay rates influence gene expression and techniques for measuring eukaryotic mRNA stability and for calculating decay constants.

Functional inactivation of genes by dominant negative mutations

TL;DR: A new approach to this problem involves the manipulation of the cloned gene to create what are known as 'dominant negative' mutations, encode mutant polypeptides that when overexpressed disrupt the activity of the wild-type gene.

Protein-protein interactions: methods for detection and analysis.

TL;DR: This review describes how binding affinities can be evaluated by techniques including protein affinity chromatography, sedimentation, gel filtration, fluorescence methods, solid-phase sampling of equilibrium solutions, and surface plasmon resonance.

Loss of a yeast telomere: Arrest, recovery, and chromosome loss

TL;DR: Yeast strains constructed in which a single telomere could be eliminated from the end of a dispensable chromosome caused a dramatic increase in loss of the chromosome in all strains examined, demonstrating that telomeres are also essential for maintaining chromosome stability.

Chromosome 21 and down syndrome: from genomics to pathophysiology.

TL;DR: Comparative genomics is beginning to identify the functional components of the chromosome and that in turn will set the stage for the functional characterization of the sequences.

A rapid alkaline extraction procedure for screening recombinant plasmid DNA

H C Birnboim, +1 more
TL;DR: In this paper, a procedure for extracting plasmid DNA from bacterial cells is described, which is simple enough to permit the analysis by gel electrophoresis of 100 or more clones per day, yet yields DNA which is pure enough to be digestible by restriction enzymes.

Arapid alkaline extraction procedure forscreening recombinant plasmid DNA

TL;DR: The method is simple enough to permit the analysis by gel electrophoresis of 100 or more clones per day yet yields plasmid DNA which is pure enough to be digestible by restriction enzymes, and achievesequate pH control without using a pH meter.

Transformation of intact yeast cells treated with alkali cations.

TL;DR: The transformation efficiency with Cs+ or Li+ was comparable with that of conventional protoplast methods for a plasmid containing ars1, although not for plasmids containing a 2 microns origin replication.

Transformation of yeast

TL;DR: This work has used recently developed hybridization and restriction endonuclease mapping techniques to demonstrate directly the presence of the transforming DNA in the yeast genome and also to determine the arrangement of the sequences that were introduced.
