Miroslawa M. Bagdasarian,Rudolf Lurz,Beate Rückert,F. C. H. Franklin,Michael Bagdasarian,Jürgen Frey,K.N. Timmis +6 more
TLDR
A cosmid that may be used for the selective cloning of large DNA fragments by in vitro lambda packaging and an analogous series of vectors that are defective in their plasmid-mobilization function, and that exhibit a degree of biological containment comparable to that of current Escherichia coli vector plasmids, are described.About:

TL;DR: Pseudomonas putida is a metabolically versatile saprophytic soil bacterium that has been certified as a biosafety host for the cloning of foreign genes.
Journal ArticleDOI
Interposon mutagenesis of soil and water bacteria : a family of DNA fragments designed for in vitro insertional mutagenesis of gram-negative bacteria
TL;DR: A series of derivatives of the omega interposon that can be used for in vitro insertional mutagenesis are constructed and insertion of these interposons in the plasmid between the promoter and the catechol 2,3-dioxygenase (C23O) gene dramatically reduced the expression of this enzyme in Escherichia coli.
Journal ArticleDOI
The Standard European Vector Architecture (SEVA): a coherent platform for the analysis and deployment of complex prokaryotic phenotypes
Rafael Silva-Rocha,Esteban Martínez-García,Belén Calles,Max Chavarría,Alejandro Arce-Rodríguez,Aitor de las Heras,A. David Páez-Espino,Gonzalo Durante-Rodríguez,Juhyun Kim,Pablo I. Nikel,Raúl Platero,Víctor de Lorenzo +11 more
TL;DR: It is argued that adoption of the SEVA format can become a shortcut to fill the phenomenal gap between the existing power of DNA synthesis and the actual engineering of predictable and efficacious bacteria.
Journal ArticleDOI
Genomic analysis of the aromatic catabolic pathways from Pseudomonas putida KT2440
TL;DR: The global view on the mineralization of aromatic compounds by P. putida KT2440 will facilitate the rational manipulation of this strain for improving biodegradation/biotransformation processes, and reveals this bacterium as a useful model system for studying biochemical, genetic, evolutionary and ecological aspects of the catabolism of aromatic compound.
Journal ArticleDOI
A versatile class of positive-selection vectors based on the nonviability of palindrome-containing plasmids that allows cloning into long polylinkers
Jeff Elhai,C P Wolk +1 more
TL;DR: A general method is described to convert virtually any small or medium-sized plasmid into a positive-selection vector and several symmetrical polylinkers and drug-resistance cassettes within the vectors.

Construction and characterization of new cloning vehicles. II. A multipurpose cloning system.
Francisco Bolívar,Raymond L. Rodriguez,Patricia J. Greene,Mary C. Betlach,Herbert L. Heyneker,Herbert W. Boyer,Jorge H. Crosa,Stanley Falkow +7 more
TL;DR: In vitro recombination techniques were used to construct a new cloning vehicle, pBR322, which is a relaxed replicating plasmid, does not produce and is sensitive to colicin E1, and carries resistance genes to the antibiotics ampicillin (Ap) and tetracycline (Tc).
Journal ArticleDOI
Construction and characterization of amplifiable multicopy DNA cloning vehicles derived from the P15A cryptic miniplasmid.
TL;DR: P15A-derived plasmids were not self-transmissible and were mobilized poorly by Hfr strains; however, mobilization was complemented by the presence of a ColE1 plasmid within the same cell.

A small cosmid for efficient cloning of large DNA fragments.
Barbara Hohn,John Collins +1 more
TL;DR: The production and use of the 6 kb cosmid pHC79, a derivative of pBR322, is described, which can be used for cloning of fragments cleaved by EcoRI, ClaI, BamHI, SalI (also XhoI and AvaI), EcaI and PstI.

Molecular and functional analysis of the TOL plasmid pWWO from Pseudomonas putida and cloning of genes for the entire regulated aromatic ring meta cleavage pathway
TL;DR: Evidence is presented that suggests the promoter operator of the meta pathway gene functions less effectively with the RNA polymerase or xylS product of E. coli than with the enzyme or product of P. putida.
