Spotiton: a prototype for an integrated inkjet dispense and vitrification system for cryo-TEM.

TLDR
A completely novel method for EM specimen preparation based on small volume (picoliter to nanoliter) dispensing using inkjet technology is described and it is demonstrated that the piezo-electric inkjet mechanism does not disrupt the structural or functional integrity of macromolecules.

Unravelling biological macromolecules with cryo-electron microscopy

TL;DR: Developments in the electron microscopy of frozen hydrated samples (cryo-electron microscopy) are providing unprecedented opportunities for the structural characterization of biological macromolecules, resulting in a wave of information about processes in the cell that were impossible to characterize with existing techniques in structural biology.
Journal ArticleDOI

Challenges and opportunities in cryo-EM single-particle analysis

TL;DR: Some of the main methods surrounding cryo-EM with an emphasis specifically on single-particle analysis are reviewed, and challenges, open questions, and opportunities for methodology development are highlighted.
Journal ArticleDOI

Cryo-electron microscopy for structural analysis of dynamic biological macromolecules.

TL;DR: The cryo-EM method can be used to determine the three-dimensional structure of biomacromolecules in near native condition at close to atomic resolution, and has the potential to reveal conformations of dynamic molecular complexes.
Journal ArticleDOI

Protein denaturation at the air-water interface and how to prevent it.

TL;DR: The structure of yeast fatty acid synthase at the air-water interface is investigated by electron cryo-tomography and single-particle image processing and it is shown that the unfolded regions face the air -water interface.

Inkjet Printing for Materials and Devices

TL;DR: Inkjet printing has been used as a free-form fabrication method for building three-dimensional parts and is being explored as a way of printing electrical and optical devices, especially where these involve organic components.

Automated molecular microscopy: the new Leginon system.

TL;DR: The primary automated data acquisition software system, Leginon, has been completely redesigned over the past two years and the system has demonstrated the capacity for high throughput data acquisition by acquiring images of more than 100,000 particles in a single session at the microscope.

The dynamics of the piezo inkjet printhead operation

TL;DR: In this paper, the physics behind the two-way coupling from the electrical to the mechanical domain through the piezoelectric actuator, where an electrical signal is transformed into a mechanical deformation of the printhead structure, is discussed.

Delivery of Human Fibroblast Cells by Piezoelectric Drop-on-Demand Inkjet Printing.

TL;DR: It is shown that the amplitude of the pulse has a small influence on cell survivability with regression analysis showing cell survival rates falling from 98% with a 40 V pulse (indistinguishable from control measurements) to approximately 94% with an 80 V pulse.
