TLDR
It is shown that after depleting interphase nuclei of histones and other nuclear proteins by treatment with nonionic detergent and high salt, intact loops of DNA can be visualized as a halo surrounding a nuclear skeleton or matrix, suggesting that the replicating DNA loops are motile with respect to their nuclear matrix anchorage sites.About:

A 3D Map of the Human Genome at Kilobase Resolution Reveals Principles of Chromatin Looping
Suhas S.P. Rao,Miriam H. Huntley,Neva C. Durand,Elena K. Stamenova,Ivan D. Bochkov,James T. Robinson,James T. Robinson,Adrian L. Sanborn,Ido Machol,Ido Machol,Arina D. Omer,Arina D. Omer,Eric S. Lander,Eric S. Lander,Eric S. Lander,Erez Lieberman Aiden +15 more
TL;DR: In situ Hi-C is used to probe the 3D architecture of genomes, constructing haploid and diploid maps of nine cell types, identifying ∼10,000 loops that frequently link promoters and enhancers, correlate with gene activation, and show conservation across cell types and species.

DNA damage activates ATM through intermolecular autophosphorylation and dimer dissociation
TL;DR: It is shown that ATM is held inactive in unirradiated cells as a dimer or higher-order multimer, with the kinase domain bound to a region surrounding serine 1981 that is contained within the previously described ‘FAT’ domain.

Apoptosis. The role of the endonuclease
TL;DR: In apoptosis, selective activation of an endogenous endonuclease appears to be responsible not only for widespread chromatin cleavage but also for the major nuclear morphologic changes, including conservation of the nucleolin-rich fibrillar center.

Mutation detection and single-molecule counting using isothermal rolling-circle amplification
Paul M. Lizardi,Xiaohua Huang,Zhengrong Zhu,Patricia Bray-Ward,David C. Thomas,David C. Ward +5 more
TL;DR: Rolling-circle amplification (RCA) driven by DNA polymerase can replicate circularized oligonucleotide probes with either linear or geometric kinetics under isothermal conditions, making it particularly amenable for the analysis of rare somatic mutations.
Journal ArticleDOI
The comet assay: a comprehensive review
TL;DR: The principles of strand break detection using both the alkaline and neutral versions of the technique are discussed, and a basic methodology with currently used variations is presented.

Chromosomal subunits in active genes have an altered conformation
Harold Weintraub,Mark Groudine +1 more
TL;DR: The results indicate that active genes are probably associated with histones in a subunit conformation in which the associated DNA is particularly sensitive to digestion by deoxyribonuclease I.

Solenoidal model for superstructure in chromatin
J T Finch,A Klug +1 more
TL;DR: It is proposed that the x-ray reflections at orders of 110 A observed in chromatin originate in the spacing between turns of the solenoid rather than that between nucleosomes along the nucleofilament.

The structure of histone-depleted metaphase chromosomes
J. R. Paulson,Ulrich K. Laemmli +1 more
TL;DR: A scaffolding model for metaphase chromosome structure is suggested in which a backbone of nonhistone proteins is responsible for the basic shape of metaphase chromosomes, and the scaffold organizes the DNA into loops along its length.

Identification of a nuclear protein matrix.
Ronald Berezney,Donald S. Coffey +1 more
TL;DR: The structural framework of the rat liver nucleus has been identified and consists of a nuclear protein matrix that is composed primarily of three acidic polypeptide fractions in the molecular weight range of 60–70,000 daltons.
