Resolving the fibrotic niche of human liver cirrhosis at single cell level
Prakash Ramachandran,Ross Dobie,John R. Wilson-Kanamori,Elena Dora,Beth E. P. Henderson,N T Luu,N T Luu,Jordan R. Portman,Kylie P. Matchett,M Brice,John A. Marwick,Richard S Taylor,Mirjana Efremova,Roser Vento-Tormo,Neil O. Carragher,Timothy J. Kendall,Jonathan A. Fallowfield,Ewen M Harrison,David R. Mole,David R. Mole,Stephen J. Wigmore,Stephen J. Marioni,John C. Marioni,Sarah A. Teichmann,Sarah A. Teichmann,Sarah A. Teichmann,Neil C. Henderson +37 more
TL;DR: Analysis of transcriptomes of more than 100,000 single human cells yields molecular definitions for non-parenchymal cell types that are found in healthy and cirrhotic human liver, and identifies markers for scar-associated macrophages and endothelial cells.
Journal ArticleDOI
Distinct Roles for CCR4 and CXCR3 in the Recruitment and Positioning of Regulatory T Cells in the Inflamed Human Liver
Ye Htun Oo,Christopher J. Weston,Patricia F. Lalor,Stuart M. Curbishley,David R. Withers,Gary M. Reynolds,Shishir Shetty,Jehan Harki,Jean C. Shaw,Bertus Eksteen,Stefan G. Hubscher,Lucy S. K. Walker,David H. Adams +12 more
TL;DR: It is proposed that CXCR3 mediates the recruitment of T Regs via hepatic sinusoidal endothelium and that CCR4 ligands secreted by DCs recruit Tregs to sites of inflammation in patients with chronic hepatitis.
Journal ArticleDOI
Platelet GPIbα is a mediator and potential interventional target for NASH and subsequent liver cancer
Mohsen Malehmir,Dominik Pfister,Suchira Gallage,Marta Szydlowska,Donato Inverso,Donato Inverso,Elena Kotsiliti,Elena Kotsiliti,Valentina Leone,Moritz Peiseler,Bas G.J. Knolle,Kristian Unger,Lars Zender,Lars Zender,Bernhard Nieswandt,Meinrad Gawaz,Achim Weber,Mathias Heikenwalder,Mathias Heikenwalder +75 more
TL;DR: It is shown that platelet number, platelet activation and platelet aggregation are increased in NASH but not in steatosis or insulin resistance, offering a potential approach to treat non-alcoholic steatohepatitis and prevent subsequent progression to hepatocellular carcinoma.
Journal ArticleDOI
Patients With Acute-on-Chronic Liver Failure Have Increased Numbers of Regulatory Immune Cells Expressing the Receptor Tyrosine Kinase MERTK
Christine Bernsmeier,Oltin T Pop,Arjuna Singanayagam,Evangelos Triantafyllou,Evangelos Triantafyllou,Vishal C. Patel,Christopher J. Weston,Stuart M. Curbishley,Fouzia Sadiq,Nikhil Vergis,Wafa Khamri,William Bernal,Georg Auzinger,Michael A. Heneghan,Yun Ma,Wayel Jassem,Nigel Heaton,David H. Adams,Alberto Quaglia,Mark Thursz,Julia Wendon,Charalambos G. Antoniades,Charalambos G. Antoniades,Charalambos G. Antoniades +23 more
TL;DR: Patients with ACLF have increased numbers of immunoregulatory monocytes and macrophages that express MERTK and suppress the innate immune response to microbes, and the number of these cells correlates with disease severity and the inflammatory response.
Journal ArticleDOI
Vascular adhesion protein-1 promotes liver inflammation and drives hepatic fibrosis
Christopher J. Weston,Emma L. Shepherd,Lee Charles Claridge,Pia Rantakari,Stuart M. Curbishley,Jeremy W. Tomlinson,Stefan G. Hübscher,Gary M. Reynolds,Kristiina Aalto,Quentin M. Anstee,Sirpa Jalkanen,Marko Salmi,David J. Smith,Christopher P. Day,David H. Adams +14 more
TL;DR: The results link the amine oxidase activity of VAP-1 with hepatic inflammation and fibrosis and suggest that targeting V AP-1 has therapeutic potential for NAFLD and other chronic fibrotic liver diseases.
